摘  要:为了考察芫花根水提物对肛瘘术后创面愈合和血管生成的影响,本研究建立了肛瘘大鼠模型并用芫花根水提物浸泡的纱布包扎大鼠伤口,共治疗14 d。给药14 d后,将大鼠肉芽组织用HE染色并在显微镜下计数毛细血管含量。通过qRT-PCR、Western blot或免疫组织化学检测肉芽组织中VEGF、VEGFR-2、COL1A1、COL3A1、TIMP-1、MMP-3、C-fos、MEK1/2、p-MEK1/2、ERK1/2和p-ERK1/2的表达。研究显示,两组大鼠治疗14 d后,与对照组相比((51.36±4.36)%),芫花根组的创面愈合率显著升高((75.35±6.37)%,p<0.05)。与对照组相比((9.26±0.87)%),芫花根组的毛细血管含量显著升高((12.45±1.16)%,p<0.05)。与对照组相比,芫花根组的VEGF和VEGFR-2的蛋白相对表达量均显著升高(p<0.05)。与对照组相比,芫花根组的COL1A1、COL3A1、TIMP-1和C-fos的mRNA和蛋白表达水平显著升高,而MMP-3显著降低(p<0.05)。与对照组相比,芫花根组的MEK1/2和ERK1/2的磷酸化水平均显著升高(p<0.05)。研究结果表明,芫花根水提物通过加速肉芽组织形成和血管生成,提高细胞外基质的功能,激活MEK/ERK信号通路来促进肛瘘术后的创面愈合。In order to investigate the effect of water extract of Daphne genkwa root on wound healing and angiogenesis after anal fistula operation,a rat model of anal fistula was established and the rat wound was wrapped with gauze soaked in water extract of Daphne genkwa root for 14 days.14 days after the administration,rat granulation tissue was stained with HE and the capillary content was counted under a microscope.The expression of VEGF,VEGFR-2,COL1 A1,COL3 A1,TIMP-1,MMP-3,C-fos,MEK1/2,p-MEK1/2,ERK1/2 and p-ERK1/2 in granulation tissue were detected by qRT-PCR,Western blotting or immunohistochemistry.Studies showed that after14 days of treatment in the two groups of rats,compared with the control group((51.36±4.36)%),the wound healing rate of the Daphne genkwa root group was significantly increased((75.35±6.37)%,p<0.05).Compared with the control group((9.26±0.87)%),the capillary content of the Daphne genkwa root group was significantly increased((12.45±1.16)%,p<0.05).Compared with the control group,the relative expression levels of VEGF and VEGFR-2 protein in the Daphne genkwa root group were significantly increased(p<0.05).Compared with the control group,the mRNA and protein expression levels of COL1A1,COL3A1,TIMP-1 and C-fos in the Daphne genkwa root group were significantly increased,while MMP-3 was significantly decreased(p<0.05).Compared with the control group,the phosphorylation levels of MEK1/2 and ERK1/2 in the Daphne genkwa root group were significantly increased(p<0.05).The water extract of Daphne genkwa root promotes wound healing after anal fistula activation by accelerating granulation tissue formation and angiogenesis,improving the function of extracellular matrix,and activating the MEK/ERK signaling pathway.
