摘　　要：目的观察MicroRNA-146b在大鼠单侧输尿管梗阻肾间质纤维化模型中的表达,探讨其在肾间质纤维化发生、发展中的意义。方法选择20只SD成年雄性大鼠,分为Sham组和UUO组。Sham组大鼠仅游离左侧输尿管,但不结扎,而UUO组大鼠结扎左侧输尿管。并分别于建模后第7天、第14天采集各组5只大鼠的左侧肾脏组织,HE染色和Masson染色评估肾间质损伤及纤维化程度,免疫组织化学检测肾组织中TGF-β1表达水平。qRTPCR检测肾组织中microRNA-146b的相对表达量。结果与相同时点Sham组相比,UUO组大鼠肾间质损伤评分、肾间质纤维化评分、肾组织microRNA-146b和TGF-β1表达水平均升高(P<0.05)。与7dUUO组大鼠相比,14dUUO组大鼠肾间质损伤评分、肾间质纤维化评分、肾组织microRNA-146b和TGF-β1表达水平升高(P<0.05)。UUO组大鼠肾组织MicroRNA-146b表达水平与肾间质损伤评分、肾间质纤维化评分、TGF-β1表达水平呈正相关(r=0.858、0.765、0.647,P<0.05)。结论UUO组大鼠肾组织microRNA-146b表达明显上调,且随着间质纤维化程度加重,表达越高。microRNA-146b可能通过参与对TGF-β1/smad信号通路的调控介导肾间质纤维化发生、发展。Objective To investigate the expression of microRNA-146 bin rat model of renal interstitial fibrosis and its significance in the occurrence and development of renal interstitial fibrosis.Methods Twenty male adult SD rats were divided into sham group and UUO group.The rats in sham group were dissociated left ureter,but not to be obstructed,and the left ureter of rats in UUO group were ligated.The left renal tissue was collected at the 7 th、14 th days after surgery.HE staining and Masson staining were used to evaluate the degree of renal interstitial damage and fibrosis.The expression of TGF-β1 in renal tissue was detected by immunohistochemistry.The relative expression of microRNA-146 bwas detected by real-time quantitative PCR.Results Compared with sham group at the correspongding time,renal interstitial injury score,renal interstitial fibrosis score,the expression of microRNA-146 band TGF-β1 in UUO group were higer than those of Sham group at the 7 th and 14 th days(P<0.05),which at 14 th day were higher than 7 th day in UUO group(P<0.05).The expression of microRNA-146 bin renal tissue was positive correlations with the renal interstitial injury score,renal interstitial fibrosis score and TGF-β1 expression level in UUO group(r=0.858,0.765,0.647,P<0.05).Conclusion The expression of microRNA-146 bis upregulated in UUO group,and the expression is higher with the aggravation of interstitial fibrosis.MicroRNA-146 bmay be involved in the regulation of TGF-β1/Smad signaling pathway and mediate the occurrence and development of renal interstitial fibrosis.
