摘 要:目的:分析不同来源野生黑果枸杞遗传差异和亲缘关系,为黑果枸杞资源鉴定和有效利用提供科学依据。方法:采用SSR分子标记对西北四省区22份野生枸杞种质材料进行鉴定,利用POPGENE 1.32和NTSYS-pc 2.10生物信息学软件对数据进行处理,分析黑果枸杞种质遗传多样性,计算遗传相似系数,绘制聚类图。结果:筛选出多态性SSR引物37对,在22个样品中共检测到233个基因位点,其中多态性位点213个,多态性比率(Polymorphism ratio,P)达91.4%。材料间遗传相似系数(Genetic similarity,GS)在0.4831~0.9364之间,平均有效等位基因数(Effective number of alleles,Ne)为1.3314,多样性指数(Nei’s gene diversity,H)为0.2106,Shannon信息指数(Shannon’s information index,I)为0.3384。聚类分析(Unweighted pair group of arithmetic means,UPGMA)显示,在相似系数为0.61和0.77处,可将22个材料分为2大类和4个亚类,不同种类的枸杞种质材料可完全分类。结论:供试枸杞种质材料间遗传多样性较为丰富,不同种类枸杞对系统聚类结果影响较大,但种内材料地域分布规律性不明显。Objective:The genetic differences and phylogenetic relationships of Lycium ruthenicum Murr.wild germplasms from Northwest desert area are analyzed in this paper,and the ues alts will provide scientific basis for identification and effective utilization of germplasm resources in the future.Methods:The genetic differences of L.ruthenicum germplasm sources was identified by SSR markers.Bioinformatics softwares POPGENE 1.32 was used to analyse the genetic diversity,and NTSYS-pc 2.10 was used to calculate the genetic similarity coefficient and draw the genetic distance tree.Results:37 pairs of primers which had clearly polymorphic bands were selected in the study.Among 233 bands from 37 pairs of primers,213 bands were polymorphic bands which occupied 91.4%of the total amplified bands.The genetic similarity range of the samples was from 0.4831 to 0.9364.On average,the numbers of effective alleles(Ne)was 1.3314,the diversity index(H)was 0.2106,the Shannon’s index(I)was 0.3384.UPGMA cluster analysis revealed that the 22 germplasm materials were divided into 2 groups with GS threshold of 0.61 and 4 subcategories with GS threshold of 0.77.Different types of L.ruthenicum can be effectively distinguish by SSR markers.Conclusion:There is rich genetic diversity in L.ruthenicum germplasms from Northwest desert area.Different types of L.ruthenicum have great influence on cluster dendrogram,but the geographical difference isn’t distinct.
