摘 要:目的:探讨柴胡桂枝汤对泛耐药鲍曼不动杆菌(pan-drug-resistant Acinetobacter baumannii,PDR-AB)侵袭的人肺癌A549细胞的保护作用及可能机制。方法:建立PDR-AB侵袭人肺癌A549细胞模型,采用柴胡桂枝汤进行干预,通过检测细胞存活率、PDR-AB的黏附率和侵袭率,细胞因子肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)的浓度以及PDR-AB生物被膜的形成能力等指标,考察柴胡桂枝汤对PDR-AB侵袭的人肺癌A549细胞的保护作用。结果:与模型组相比,柴胡桂枝汤组对PDR-AB侵袭的人肺癌A549细胞有保护作用,可显著提高人肺癌A549细胞存活率(30、15及7.5 mg/ml,P <0.05);显著抑制PDR-AB对A549细胞的黏附作用(柴胡桂枝汤浓度为7.5 mg/ml时,P <0.05;柴胡桂枝汤浓度为30、15 mg/ml时,P <0.01);显著抑制PDR-AB对A549细胞的侵袭作用(柴胡桂枝汤浓度为30 mg/ml时,P <0.05);显著抑制PDR-AB生物被膜的形成(柴胡桂枝汤浓度为15 mg/ml时,P <0.05;柴胡桂枝汤浓度为30 mg/ml时,P <0.01);显著降低TNF-α和IL-6的释放水平(柴胡桂枝汤浓度为30、15 mg/ml时,P <0.01),上述差异均有统计学意义。结论:柴胡桂枝汤对PDR-AB侵袭的人肺癌A549细胞具有一定的保护作用。OBJECTIVE:To probe into the protective effect of Chaihu Guizhi decoction on pan-drug-resistant Acinetobacter baumannii(PDR-AB)invading human lung cancer A549 cells and its possible mechanism.METHODS:A model of PDR-AB invading human lung cancer A549 cells was established,and Chaihu Guizhi decoction was used to conduct intervention.Through detecting cell survival rate,PDR-AB adhesion rate and invasion rate,concentrations of tumor necrosis factorα(TNF-α)and interleukin 6(IL-6)concentration,forming ability of PDR-AB biofilm and other indicators,and the protective effect of Chaihu Guizhi decoction on PDR-AB invading human lung cancer A549 cells was inspected.RESULTS:Compared with the model group,Chaihu Guizhi decoction had protective effect on PDR-AB invading human lung cancer A549 cells,which can significantly improve the survival rate of human lung cancer A549 cells(30,15 and 7.5 mg/ml,P<0.05);significantly inhibit the adhesion of PDR-AB to A549 cells(P<0.05 when the concentration of Chaihu Guizhi decoction was 7.5 mg/ml;P<0.01 when the concentration of Chaihu Guizhi decoction was 30 and 15 mg/ml);significantly inhibit the invasion effect of PDR-AB on A549 cells(P<0.05 when the concentration of Chaihu Guizhi decoction was 30 mg/ml);significantly inhibit the formation of PDR-AB biofilm(P<0.05 when the concentration of Chaihu Guizhi decoction was 15 mg/ml;P<0.01 when the concentration of Chaihu Guizhi decoction was 30 mg/ml);significantly reduce the release levels of TNF-αand IL-6(P<0.01 when the concentration of Chaihu Guizhi decoction was 30 and 15 mg/ml),with statistically significant differences.CONCLUSIONS:Chaihu Guizhi decoction has a certain protective effect on PDR-AB invading human lung cancer A549 cells.
