摘 要:目的:探讨白藜芦醇对慢性青光眼大鼠视神经保护作用及对B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)、含半胱氨酸的天冬氨酸蛋白水解酶-9(Caspase-9)表达的影响。方法:选取清洁级SD大鼠100只,按照随机数字表法分为对照组、模型组和白藜芦醇低、中及高剂量组(20、40及80 mg/kg),每组20只。模型组和白藜芦醇低、中及高剂量组大鼠建立慢性青光眼模型,建模成功后,白藜芦醇低、中及高剂量组予相应药物腹腔灌胃,持续12周,模型组及对照组大鼠给予相应体积的0.9%氯化钠溶液。实验结束后测量五组大鼠眼压、视网膜细胞凋亡水平,观察大鼠视网膜病理结构变化,测定视网膜Bcl-2、Bax、Caspase-9 mRNA和蛋白水平。结果:与对照组比较,模型组大鼠眼压、视网膜神经节细胞凋亡率,Bax、Caspase-9 mRNA和蛋白水平明显升高,视网膜Bcl-2 mRNA和蛋白水平明显降低,差异均有统计学意义(P <0.05);与模型组比较,白藜芦醇低、中及高剂量组大鼠眼压、视网膜神经节细胞凋亡率,Bax、Caspase-9 mRNA蛋白水平明显降低,视网膜Bcl-2 mRNA和蛋白水平明显升高,差异均有统计学意义(P <0.05),且呈剂量依赖性。对照组大鼠视网膜神经结构正常;模型组大鼠的高眼压导致视网膜神经死亡,核固缩,且有大量炎症细胞浸润;白藜芦醇低、中及高剂量组大鼠视神经纤维排列无规律,纤维间质少见水肿与空泡,伴随少量炎症细胞浸润。结论:白藜芦醇能降低慢性青光眼大鼠的眼压,对视网膜神经节细胞具有保护作用;其机制与白藜芦醇能明显抑制视网膜神经节细胞Bax、Caspase-9 mRNA和蛋白的表达,提高Bcl-2 mRNA和蛋白的表达水平,进而抑制视网膜神经节细胞的凋亡有关。OBJECTIVE: To probe into the protective effect of resveratrol on optic nerve and its effect on the expression of B lymphocytoma-2 gene( Bcl-2),Bcl-2 associated X protein( Bax) and cysteinyl aspartate specific proteinase-9( Caspase-9) in rats with chronic glaucoma. METHODS: Totally 100 clean-grade SD rats were selected and divided into control group,model group,low,medium and high dose groups of resveratrol( 20,40 and 80 mg/kg)via random number table,with 20 rats in each group. Rats in the model group and low,medium and high doses of resveratrol groups were collected to establish chronic glaucoma models,the low,medium and high doses groups of resveratrol were given intraperitoneal gavage with corresponding drugs for 12 weeks after the modeling was successful,while the model group and control group were given corresponding volume of 0. 9% sodium chloride solution. After the test,the intraocular pressure and retinal cell apoptosis levels of the five groups of rats were measured,the pathological structure changes of rat retina were observed,the mRNA and protein levels of Bcl-2,Bax and Caspase-9 in the retina were measured. RESULTS: Compared with the control group,the intraocular pressure,apoptosis rates of retinal ganglion cell,mRNA and protein levels of Bcl-2,Bax and Caspase-9 in the model group of resveratrol had been significantly increased, while the mRNA and protein levels of Bcl-2 in retina had been significantly decreased,with statistically significantly differences( P < 0. 05);compared with the model group,the intraocular pressure,apoptosis rates of retinal ganglion cell,mRNA and protein levels of Bcl-2,Bax and Caspase-9 in low,medium and high dose groups of resveratrol had been significantly decreased,while the mRNA and protein levels of Bcl-2 in retina had been significantly increased,with statistically significantly differences( P < 0. 05),which presented a dose-dependent.
