摘 要:目的:了解不同炮制方法对曼陀罗子饮片中阿托品和东莨菪碱含量的影响,保证用药安全有效。方法:运用传统炮制方法甘草制法和醋制法对新疆曼陀罗子生药材进行炮制。以阿托品和东莨菪碱含量变化为考察指标,采用高效液相色谱法进行含量测定,色谱条件为Super C(18)柱(4.6 mm×250 mm,5μm),流动相为甲醇-水(V∶V=30∶70,水中含0.02 mol/L醋酸钠溶液、0.02%三乙胺,用醋酸调节p H至6.0),流速为1.0 ml/min,检测波长为215 nm,柱温为室温(25℃),进样量为10μl。结果:醋制曼陀罗子中东莨菪碱的平均含量为1.084 mg/g,阿托品为4.521 mg/g。甘草制曼陀罗子中东莨菪碱的含量为1.098 mg/g,阿托品为3.293 mg/g。结论:经过综合对比,最终确定甘草制曼陀罗子的减毒效果优于醋制法。OBJECTIVE:To investigate the effects of different processing methods on content determination of atropine and scopolamine in different processed products of Datura stramonium L.,so as to ensure the safety and effectiveness of medication.METHODS:The traditional processing methods of glycyrrhiza and vinegar were used to process the raw materials of Datura stramonium L.in Xinjiang.The content changes of atropine and scopolamine were used as the study indexes,the content was determined by high performance liquid chromatography(HPLC).The chromatographic conditions were Super C 18 column(4.6 mm×250 mm,5μm),the mobile phase was methanol-water(V∶V=30∶70,the water contained 0.02 mol/L sodium acetate solution and 0.02%triethylamine,pH was adjusted to 6.0 with acetic acid).The flow rate was 1.0 ml/min,the detection wavelength was 215 nm,the column temperature was room temperature(25℃),and the sample injection volume was 10μl.RESULTS:The average content of scopolamine and atropine in Datura stramonium L.by glycyrrhiza was 1.084 mg/g and 4.521 mg/g respectively.The content of scopolamine and atropine in Datura stramonium L.by vinegar was 1.098 mg/g and 3.293 mg/g respectively.CONCLUSIONS:Through the comprehensive comparison,it is determined that the detoxification effects of Datura stramonium L.by glycyrrhiza is better than that of vinegar.
