摘  要:类风湿关节炎(RA)是一种以对称性、多关节炎为主要临床表现的自身免疫性疾病,属于临床常见的慢性难治性疾病。若不进行及时有效的治疗,可导致关节破坏及内脏损伤。RA发病机制复杂,涉及多条信号通路,寻找有效的靶点对RA的治疗有重要意义。沉默信息调节因子1可以作用于多个与RA相关的信号通路,如核因子κB通路、激活蛋白-1通路、缺氧诱导因子1α通路、促分裂原活化的蛋白激酶通路等,在调控炎症反应发生、微血管生成和关节软骨破坏等方面发挥重要作用,可能是RA的潜在治疗靶点。Rheumatoid arthritis(RA)is a kind of chronic systemic autoimmune disease characterized by symmetry and polyarthritis.It is a common chronic refractory disease in clinic.If not treated in time,it can often lead to joint damage and internal injury.The pathogenesis of RA is complex and involves many signaling pathways,and finding effective targets has important practical significance for the treatment of RA.Silent information regulator type 1 can act on multiple RA related signaling pathways,such as nuclear factor-κB pathway,activator protein-1 pathway,hypoxia-inducible factor-1αpathway,mitogen-activated protein kinases pathway,which plays an important role in the regulation of inflammation,the generation of microvessels and the destruction of articular cartilage,and may be a potential therapeutic target for RA.
