摘 要:目的研究相反药对甘遂-甘草的大鼠血浆代谢组学,以探讨其"相反"的作用机制。方法实验大鼠分别给予生理盐水及甘遂-甘草(euphorbia kansui-glycyrrhiza,GS-GC)提取物,连续灌胃14 d,取大鼠肝脏组织进行切片观察,并同时测定大鼠血清谷丙转氨酶、谷草转氨酶等生化指标以评价甘遂-甘草对大鼠的肝毒性。应用UHPLC-FT-ICR-MS技术对大鼠血浆样品进行分析。采用多元统计分析,建立OPLS-DA模型,结合HMDB、KEGG等数据库确定甘遂-甘草与空白组之间的差异性代谢物。结果甘遂-甘草组大鼠出现肝损伤,且与空白组得到良好区分,共找到7个内源性血浆生物标记物。结论相反药对甘遂-甘草对大鼠造成了一定的肝损伤,在一定程度上干扰了磷脂代谢和氨基酸代谢。Objective To study the rat plasma metabonomics of Gansui-gancao and explore the mechanism of the incompatible herb pair.Methods Rats were given normal saline and Gansui-gancao(Euphorbia kansui-Glycyrrhiza,GS-GC) extracts for 14 days.Liver tissues histopathology examinations and serum biomedical indicators alanine aminotransferase(ALT) and aspartate aminotransferase(AST) detection were carried out for the assessment of the liver toxity of Gansui-gancao.UHPLC-FT-ICR-MS technology was used for plasma analysis.Multivariate statistical analysis was used to build an OPLS-DA model.HMDB and KEGG database were used for identify the biomarkers between GS-GC and Con group.Results Rats in GS-GC group had liver injury,besides,GS-GC and Con group were separated well.A total of 7 metabolites were found as biomarkers.Conclusion Incompatible herb pair Gansui-Gancao induced liver injury and disturbed phospholipid metabolism and phenylalanine metabolism of rats in some degree.
