摘  要:传统观念认为,成体动物的神经系统是极为稳定且不可再生的。而近几十年的研究表明,成体啮齿类及灵长类动物海马、侧脑室下区等部位可不断产生新的神经元,打破了原有对神经干细胞的错误理解。已有证据显示海马神经发生参与了多种中枢神经系统疾病,如阿尔茨海默病、精神分裂症以及抑郁症等的病理进程。而近期研究发现,药物成瘾同样与海马神经发生高度相关,一方面表现为成瘾性药物能显著影响海马神经干细胞的增殖、分化及成熟;而另一方面,促进海马神经发生则可能通过干预成瘾记忆进而抑制多种成瘾性药物导致的成瘾行为。本文主要针对海马神经发生在药物成瘾中所发挥的作用及相关机制做一综述,以期为药物成瘾的治疗提供新的策略。It has been traditionally believed that the central nervous system(CNS)of adult animals is non-renewable.However,studies in recent decades have indicated that the brains of adult rodents and primates can constantly generate new neurons in the nich⁃es such as subgranular zone and subventricular zone,thus breaking the original misunderstanding of neural stem cells.Evidence has also showed that neurogenesis may participate in regulating the pathological processes of various central system diseases such as Alzheim⁃er's disease(AD),schizophrenia,and depression.Moreover,recent studies have found that drug addiction is highly associated with hippocampal neurogenesis.On the one hand,the addictive drugs can significantly influence the proliferation,differentiation and matu⁃ration of hippocampal neural stem cells.On the other hand,promoting the hippocampal neurogenesis has been shown to inhibit drug addiction by interfering with the addiction memory.In the present review,we focus on the role of neurogenesis in drug addiction as well as the related molecular mechanism,to provide novel therapeutic approaches to the management of drug addiction.
