摘 要:目的采用高效液相色谱法,建立同时测定荷叶中N-去甲基荷叶碱、O-去甲基荷叶碱、荷叶碱、莲碱4种生物碱含量的一测多评法。方法采用高效液相色谱法,荷叶碱为内标,分别建立荷叶碱与N-去甲基荷叶碱、O-去甲基荷叶碱、莲碱的相对校正因子,计算荷叶中N-去甲基荷叶碱、O-去甲基荷叶碱、莲碱的含量,实现一测多评。同时采用外标法测定荷叶中4种生物碱类成分的量,并将计算结果与一测多评法进行比较。色谱柱为Phenomenex C18(250 mm×4.6 mm,5μm);流动相为乙腈-水-三乙胺-冰醋酸(49∶49.2∶1.25∶0.48);检测波长为270 nm;流速为1 mL/min;柱温为30℃;进样量为10μL。结果荷叶中的N-去甲基荷叶碱、O-去甲基荷叶碱、荷叶碱、莲碱分别在0.0209~0.4184μg(r=1.0000)、0.0100~0.1992μg(r=1.0000)、0.0197~0.3936μg(r=0.9999)、0.0078~0.1568μg(r=1.0000)范围内呈良好的线性关系;平均回收率(n=6)分别为101.16%、98.73%、101.06%、95.95%,RSD值分别为1.93%、1.75%、1.33%、2.00%。结论以荷叶碱为内标同时测定N-去甲基荷叶碱、O-去甲基荷叶碱、莲碱的一测多评法可用于荷叶的定量分析。Objective:To establish a quantitative analysis of multi-components by single marker method for simultaneous determination of four alkaloids contents of N-nornuciferine,O-nornuciferine,nuciferinea and roemerine in Nelumbinis Folium by using HPLC.Methods:With nuciferinea as an internal standard,the relative correction factors of N-nornuciferine,O-nornuciferine and roemerine were calculated respectively,and the contents of N-nornuciferine,O-nornuciferineand roemerine in Nelumbinis Folium were determined to achieve multiple evaluation by using HPLC.At the same time,the quantity of four alkaloids in Nelumbinis Folium was measured by external standard method,and the difference between calculated value and measured value was compared,so as to verify the scientific nature and feasibility of multi-evaluation method.The chromatographic separation was carried on Phenomenex C18 column(250 mm×4.6 mm,5μm)in isocratic elution with the mobile phase of acetonitrile-water-triethylamine-acetic acid(49:49.2:1.25:0.48).The detection wavelength was 270 nm;the flow rate was set at 1 mL/min;The column temperature was set at 30℃;the sample size was 10μL.Results:At concentrations ranging between 0.0209-0.4184μg(r=1.0000),0.0100-0.1992μg(r=1.0000),0.0197-0.3936μg(r=0.9999),0.0078-0.1568μg(r=1.0000),peak area of N-nornuciferine,O-nornuciferine,nuciferinea and roemerineshowed good linear relationship,and the average recoveries(n=6)were 101.16%(RSD 1.93%),98.73%(RSD 1.75%),101.06%(RSD 1.33%),95.95%(RSD 2.00%)respectively.Conclusion:The method of simultaneous determination of N-nornuciferine,O-nornuciferine and roemerine with nuciferinea as internal standard by single marker can be used for quantitative analysis of Nelumbinis Folium.
