摘 要:目的评价LC-MS/MS法测定人血浆中肌苷浓度的不确定度。方法分析LC-MS/MS法测定人血浆中肌苷浓度的整个过程,从精密度、称量、样品配制、生物样品处理过程、基质效应、液质联用仪、线性拟合等方面确定不确定度的来源及其对结果影响的大小,并计算各分量不确定度,最后合成并扩展。结果人血浆中低浓度(62.8 ng/ml)、中浓度(502.5 ng/ml)、高浓度(2010 ng/ml)肌苷的扩展不确定度分别为17.36,90.88,357.46 ng/ml(k=2,P=95%)。天平称量引入的不确定度为0.025,线性拟合引入的不确定度为0.123(低浓度质控样品)、0.0145(中浓度质控样品)及0.00478(高浓度质控样品)。结论LC-MS/MS法测定人血浆中肌苷浓度的不确定度主要由天平称量和线性拟合引入。Objective To evaluate the uncertainty for determination of inosine in human plasma by LC-MS/MS.Methods Uncertainty sources were established and evaluated in the whole process during the determination of inosine,including effect of precision,balance weighing,sample preparation,processing of plasma samples,matrix effect,LC-MS/MS,fitting of standard curve,etc.Finally,the combined and expanded uncertainties were calculated.Results The expanded uncertainties of low dose(62.8 ng/ml),medium dose(502.5 ng/ml)and high dose(2010 ng/ml)of inosine in human plasma were 17.36,90.88,357.46 ng/ml,respectively(k=2,P=95%).The uncertainty of balance weighing was 0.025,and the uncertainty of fitting of standard curve was 0.123(low dose),0.0145(medium dose)and 0.00478(high dose),respectively.Conclusion The uncertainty of the determination of inosine by LC-MS/MS is mainly caused by balance weighing and fitting of standard curve.
