摘 要:目的:探讨杨梅素对大鼠脊髓损伤(SCI)的作用及机制。方法:将雄性SD大鼠随机分为6组,即假手术组、模型组、杨梅素低剂量组、杨梅素中剂量组、杨梅素高剂量组和阳性对照组,每组10只。除假手术组外,其他各组采用Allen’s法建立SCI大鼠模型。比较各组大鼠脊髓湿/干重比,采用Basso-Beattie-Bresnahan(BBB)运动评分表评估大鼠运动功能,酶联免疫吸附测定(ELISA)法检测大鼠脊髓组织白细胞介素(IL)-1β、IL-6和肿瘤坏死因子(TNF)-α含量,苏木精-伊红染色和尼氏染色观察大鼠脊髓组织病理变化,Western blotting法检测大鼠脊髓组织磷酸化(p)-IκBα/IκBα、p-p65/p65、p-p38/p38、磷酸化细胞外调节蛋白激酶(p-ERK)/ERK、磷酸化c-Jun氨基末端激酶(p-JNK)/JNK蛋白表达水平。结果:与模型组比较,杨梅素中、高剂量组及阳性对照组BBB评分显著升高,脊髓湿/干重比和IL-6、IL-1β、TNF-α含量显著降低,神经元坏死和水肿减少,尼氏体数量显著增加(均P<0.05)。与模型组比较,杨梅素高剂量组和阳性对照组p-IκBα/IκBα、p-p65/p65、p-p38/p38、p-ERK/ERK、p-JNK/JNK比值显著降低(均P<0.05)。结论:杨梅素能够促进SCI大鼠运动功能恢复,减少炎症反应和神经元凋亡,其作用可能与抑制NF-κB和MAPK信号通路有关。Objective:To explore the effect and mechanism of myricetin on spinal cord injury(SCI).Methods:Male SD rats were randomly divided into 6 groups:sham-operation group,model group,myricetin low-dose group,middle-dose group,high-dose group and positive control group,10 rats in each group.SCI rat models were established by Allen’s method in all groups except for the sham-operation group.The spinal cord wet/dry weight ratio was compared.The Basso-Beattie-Bresnahan(BBB)scale was used to evaluate motor function of rats.Enzyme linked immunosorbent assay(ELISA)was used to detect the contents of interleukin(IL)-6,IL-1βand tumor necrosis factor(TNF)-αin spinal cord tissues.Hematoxylin-eosin(HE)staining and Nissl staining were perform to observe pathological changes of spinal cord tissues.Western blotting was used to determine p-IκBα/IκBα,p-p65/p65,p-p38/p38,p-ERK/ERK,p-JNK/JNK protein expression levels.Results:Compared with the model group,the BBB score and the number of Nissl bodys in the middle-dose group,high-dose group and positive control group were increased,while the wet-to-dry weight ratio of spinal cord,the levels of IL-6,IL-1β and TNF-α as well as neuronal necrosis and edema were decreased significantly(P<0.05).Compared with the model group,the ratios of p-IκBα/IκBα,p-p65/p65,p-p38/p38,p-ERK/ERK,and p-JNK/JNK were significantly reduced in the high-dose group and positive control group(P<0.05).Conclusion:Myricetin can promote the recovery of motor function,reduce inflammatory response and neuronal apoptosis in SCI rats,and its effect may be related to the inhibition of NF-κB and MAPK signaling pathways.
