摘  要:目的:探讨鼠胚神经干细胞联合鼠神经生长因子治疗颈脊髓压迫大鼠的效果及机制。方法:选择雄性Sprague Dawley(SD)大鼠建立颈脊髓压迫模型,成模后随机分为3组:模型组(颈静脉注射等量生理盐水)、常规治疗组(颈静脉注射500U/kg·d^-1鼠神经生长因子)与联合组(在常规治疗组基础上给予颈静脉注射60μL鼠胚神经干细胞)。然后分别采用Basso-Beattie-Bresnahan(BBB)运动功能评分法评定大鼠的肢体运动功能;流式细胞仪检测细胞凋亡指数;DAPI染色观察神经元形态并计算正常神经元细胞比例;Western blot法检测STAT-5、BCL-2蛋白表达。结果:治疗前1 d,3组BBB评分结果比较,差异无统计学意义(P>0.05);常规治疗组与联合组治疗后7 d与14 d的BBB评分均显著高于模型组,联合组也显著高于常规治疗组(P<0.05)。常规治疗组与联合组治疗后14 d的神经元细胞凋亡指数显著低于模型组,联合组也显著低于常规治疗组(P<0.05);正常神经元比例高于模型组,联合组则较常规治疗组显著降低(P<0.05);STAT-5、BCL-2蛋白相对表达水平高于模型组,联合组也显著高于常规治疗组(P<0.05)。结论:鼠胚神经干细胞联合鼠神经生长因子治疗颈脊髓压迫大鼠能促进提高肢体功能,促进STAT-5与BCL-2蛋白的表达,减少神经元细胞凋亡,提高正常神经元比例。Objective:To investigate the effects and mechanism of rat embryonic neural stem cells combined with rat nerve growth factor on cervical spinal cord compression in rats.Methods:Male Sprague Dawley(SD)ratswere used to establish models of cervical spinal cord compression,and then randomly divided into three groups:model group(an equal amount of normal saline injected into the jugular vein),conventional treatment group(jugular vein injection of 500 U/kg·d^-1 mouse nerve growth factor)and combination group(jugular vein injection of 60μL mouse embryonic neural stem cells on the basis of conventional treatment group).The BBB score was used to evaluate the motor function of the rats'limbs,the flow cytometry was used to detect apoptosis index,DAPI staining was used to observe neuron morphology and the proportion of normal neuron cells was calculated.Western blot method was used to detect the expression of STAT-5 and BCL-2 proteins.Results:One day before treatment,there was no significant difference in BBB scores among the three groups(P>0.05).The BBB score in the conventional treatment group and the combination group was significantly higher than that in the model group on 7 and 14 days after treatment,and the combination group was significantly higher(P<0.05).The apoptosis index of neurons in the conventional treatment group and the combination group was significantly lower than that in the model group on day 14 after treatment,and the combination group was significantly lower(P<0.05).The proportion of normal neurons was higherin the conventional treatment group and the combination group than that in the model group,and the combination group was significantly lower(P<0.05).The relative expression levels of STAT-5 and BCL-2 protein were higher than those in the model group,and the combination group was significantly higher(P<0.05).Conclusion:Rat embryonic neural stem cells combined with rat nerve growth factor in the treatment of cervical spinal cord compression rats can improve limb function,elevate the expression of S
