摘 要:为了准确鉴定采自黑龙江省自然感染犬小肠内的两种吸虫,在对虫体进行形态学初步鉴定后,采取PCR方法扩增两种虫体的核糖体内转录间隔区(ITS)序列,并与棘口科相关序列进行比对和进化分析。结果表明:两种吸虫的ITS序列长度分别为1233 bp和1046 bp。序列分析发现虫体A与GenBank上来自越南的日本棘隙吸虫(KT873311.1)ITS2序列同源性为99.5%;吸虫B与来自日本的圆圃似颈吸虫(圆圃棘口吸虫的同物异名AB189982.1)和中国的圆圃棘口吸虫(KX832896.1)同源性均为100%。基于ITS2序列的进化分析显示棘口科吸虫形成两个大的分支,棘隙属与冠孔属为一分支,其它属虫体形成另一分支。虫体A与5条日本棘隙吸虫,虫体B与圆圃似颈吸虫和圆圃棘口吸虫分别形成独立分支,亲缘关系最近。因此鉴定这两种吸虫分别为日本棘隙吸虫和圆圃似颈吸虫。In order to identify the species of parasites collected from a dog in Heilongjiang province,the morphological identification of two worms were occurred,and the internal transcribed spacer(ITS)sequences of the nuclear ribosomal DNA were amplified by PCR.ITS sequences of two parasites were compared with the related sequences of Echinostomatidae trematodes available in GenBank and the phylogenetic relationships were inferred.The results showed that the length of ITS sequences of trematodes A and B were 1233 bp and 1046 bp,respectively.Sequence analysis showed that the identity of ITS2 sequences between trematode A and Ec-hinochasmus japonicus(KT873314)form Vietnam available in GenBank was 99.5%,the identity of ITS2 sequences among trematode B,Isthmiophora hortensis(AB189982.1,synonym Echinostoma hortense)form Japan and E.hortense form China(KX832896.1)were all 100%.Evolutionary analysis based on ITS2 sequences showed that Echinostomatidae trematodes are divided into two large branches,Echinochasmus and Stephanoprora formed one clade,and other genera formed another clade.Trematode A was clustered together with five E.japonicus,and trematode B was clustered together with I.hortensis and E.hortense available in GenBank,respectively.So,these two trematodes were identified as E.japonicus and I.hortensis,respectively.
