摘  要:建立了一种高动能研磨结合超高液相色谱-串联质谱(UPLC-MS/MS)法快速检验毛发中常见毒品及其代谢物的方法。考察了提取剂种类及研磨方式等因素对检验结果的影响,确定了最佳实验方法,20 mg头发置于专用研磨管内,加入1 mL甲醇,振荡混匀,在3000 r/min的频率下研磨3 min,超声2 h,离心过膜进样。毛发中7种毒品及其代谢物含量在0. 05~5 ng/mg的范围内线性关系良好,相关系数r均大于0. 99,回收率在63. 1%~99. 5%之间,最小检出限5 pg/mg,能够满足基层公安工作需要。通过分析上百份吸毒人员的毛发样本,确定了毒品含量分布规律及甲基苯丙胺吸食者毛发中苯丙胺与甲基苯丙胺的含量比率同甲基苯丙胺的含量呈负相关的关系,为禁毒人员提供理论支撑与方法参考。A rapid and sensitive method of high kinetic energy grinding and ultra-performance liquid chromatography coupled with mass spectrometry( UPLC-MS/MS) was developed for simultaneously determining common drugs and metabolites in hair. The effects of the extracting agent type and the grinding way were investigated,and the best experimental scheme was determined. The 20 mg hair was placed in a special grinding bottle,and then 1 m L methanol was added and mixed in an oscillating manner. The hair was grounded for 3 min at the frequency of 3000 r/min. The grinding fluid was disposed by ultrasonic for 2 hours and detected after centrifugalization and filtration. The contents of seven kinds of drugs and metabolites in hair were in good linear in the range of 0. 05-5 ng/mg,with their coefficients higher than 0. 99. The extraction recoveries ranged from63. 1% to 99. 5% for all of the analytes,and their limits of detection( LODs) were 5 pg/mg,which could meet the needs of grassroots public security work. Meanwhile,hundreds of hair samples of drug addicts were tested by this method. The distribution of drug content was determined and the content ratio of amphetamine and methamphetamine was carried on the statistical analysis,which was proved to be basic negatively correlated with the content of methamphetamine. These analyses and conclusions can provide theoretical support and method reference for the drug-related cases.
