摘 要:以4份百脉根品系成熟叶片为材料,采用石蜡制片法和光学显微技术测定叶片上表皮厚度、叶肉厚度、叶脉厚度等10个与抗旱性相关的解剖结构特征参数并进行分析评价,以期筛选出抗旱性强的百脉根新品系。结果表明:不同品系的叶片上表皮厚度、叶片厚度、叶肉厚度、叶脉厚度、海绵组织厚度、栅栏组织厚度、栅海比、叶片栅栏组织结构紧密度、叶片海绵组织结构疏松度均表现出极显著差异(P<0.01),变异系数最大的是叶片上表皮厚度,为24.98%,最小的是叶片下表皮厚度,为2.86%;性状间相关分析表明,叶片下表皮厚度与栅海比显著负相关(P<0.05),叶片厚度与叶肉厚度、栅栏组织厚度、海绵组织厚度之间呈极显著或显著相关(P<0.01或P<0.05),叶肉厚度与栅栏组织厚度、海绵组织厚度显著正相关(P<0.05);主成分分析表明,前3个主成分的特征值总和为10.001,累计贡献率达100%,能概括10个指标的主要信息;隶属函数法综合评价表明,百脉根新品系B1抗旱性最强,品系B4抗旱性最差。In order to screen out the drought-resistant Lotus corniculatus strains,four mature leaves of Lotus corniculatus strains were used as materials,and ten anatomical structure parameters related to drought resistance,such as leaf epidermis thickness,mesophyll thickness and vein thickness,were measured by paraffin section method and optical microscopy technology.The results showed that:The thickness of leaf epidermis,leaf,mesophyll,vein,spongy tissue,palisade tissue,ratio of palisade to spongy,compactness of palisade tissue and porosity of spongy tissue were significantly different among different strains(P<0.01).The highest coefficient of variation was the leaf epidermis thickness of leaves(24.98%),and the lowest was leaf subcutaneous thickness(2.86%).Correlation analysis showed that there were significant correlation or extreme significant correlation between leaf lower epidermis thickness and ratio of palisade to spongy,between leaf thickness and mesophyll thickness,palisade tissue thickness and sponge tissue thickness.Principal component analysis showed that the sum of the eigenvalues of the first three principal components was 10.001,and the cumulative contribution rate reached 100%,which could summarize the main information of the 10 indicators.The comprehensive evaluation of drought resistance by membership function method showed that the drought resistance of the new strain B1 was the strongest,and the strain B4 was the worst.
