摘 要:目的:研究脑缺血大鼠给予脑泰方(NTF)干预后对脂质代谢蛋白表达的影响。方法:将44只大鼠随机分为Sham组、MCAO组、MCAO+NTF组和MCAO+Fer-1组。采用大脑中动脉栓塞(MCAO)模型,术后12h采用Longa神经行为学评分标准记录大鼠活动;丙二醛(MDA)测定检测脑缺血部位脂质过氧化水平;Western blot及RT-PCR检测各组脑组织谷胱甘肽过氧化物酶4(GPX4)、长链脂酰辅酶A合成酶4(ACSL4)、磷脂胆碱酰基转移酶3(LPCAT3)蛋白及基因表达;ROS检测各组脑组织活性氧(ROS)水平。结果:与MCAO组比较,MCAO+NTF组的神经行为学评分下降(P<0.05),GPX4蛋白及mRNA表达升高(P<0.05,P<0.01),LPCAT3、ACSL4蛋白及mRNA表达下调(P<0.05,P<0.01),MDA、ROS水平降低(P<0.01)。结论:脑泰方可以上调GPX4蛋白和基因的表达,下调ACSL4、LPCAT3蛋白和基因的表达,抑制不饱和脂肪酸的氧化,降低脂质过氧化水平和ROS的沉积,从而改善脑缺血后的神经损伤。Objective:To investigate the effects of Naotaifang(NTF)on lipid metabolism protein expression in cerebral ischemia rats.Methods:Forty-four SD rats were randomly divided into Sham operation group,Model group(MCAO group),Model+Naotaifang group(MCAO+NTF group)and Model+Ferrostatin-1 group(MCAO+Fer-1 group).After cerebral ischemia for 12 hours,the behavioral activity of rat was recorded by Longa’s neurobehavioral score standard;Detecting the lipid peroxidation level in ischemic area of rats in each group by MDA;The expressions of glutathione peroxidase 4(GPX4),long-chain-fatty-acidCoA ligase 4(ACSL4),phosphatidylcholine acyltransferase 3(LPCAT3)proteins and genes were detected by Western blot and RT-PCR;Detecting the reactive oxygen species(ROS)level in ischemic area of rats in each group by fluorescent.Results:Compared with MCAO group,the Longa’s neurobehavioral score was significantly decreased(P<0.05),the protein and mRNA expressions of GPX4 were significantly increased(P<0.05,P<0.01),the protein and mRNA expressions of ACSL4 and LPCAT3 were significantly decreased(P<0.05,P<0.01),levels of MDA and ROS were significantly reduced(P<0.01).Conclusion:Naotaifang can increased the protein and gene expressions of GPX4,reduce the protein and gene expressions of ACSL4 and LPCAT3,inhibit oxidation of unsaturated fatty acids and reduce lipid peroxidation level and ROS deposition.Thus,it can decrease nerve damage after cerebral ischemia.
