摘 要:目的:探讨糖肾宁含药血清对于高糖刺激下足细胞上皮间质转分化的影响。方法:将条件永生小鼠足细胞系分为正常对照组,高糖组,糖肾宁含药血清低、中、高剂量组,CCK-8法检测各组足细胞细胞活力,倒置显微镜观察各组足细胞细胞形态,鬼笔环肽-罗丹明染色观察各组足细胞细胞骨架变化,免疫荧光染色及Western blot法检测各组足细胞P-钙粘蛋白(P-cadherin)、成纤维细胞特异蛋白-1(FSP-1)蛋白表达水平。结果:与正常对照组比较,高糖组足细胞细胞活力降低(P<0.05),细胞形态变化及细胞骨架重构,P-cadherin蛋白表达下调(P<0.05),FSP-1蛋白表达上调(P<0.05);与高糖组比较,糖肾宁含药血清组足细胞细胞活力升高(P<0.05),细胞形态变化及细胞骨架重构减轻,P-cadherin蛋白表达上调(P<0.05),FSP-1蛋白表达下调(P<0.05)。结论:糖肾宁含药血清可以减轻高糖刺激引起的足细胞上皮间质转分化。Objective:To observe the effects of drug-containing serum of Tangshenning(TSN)on podocyte EMT in high-glucose environment.Methods:Differentiated mouse podocytes in vitro were divided into following groups:normal control group,high glucose group,low,medium and high dose drug-containing serum of TSN groups.Cell viability was detected by CCK-8 assay.Cell morphology was observed by microscope and rhodamine staining.Immunofluorescence and Western blot were used for detecting protein expression of P-cadherin and FSP-1 in our study.Results:Compared with normal control group,cell viability of high glucose group was significantly decreased(P<0.05),cell morphological changes and cytoskeleton remodeling,P-cadherin protein expression was significantly decreased(P<0.05),FSP-1 protein expression was significantly increased(P<0.05).Compared with high glucose group,cell viability of drug-containing serum of TSN group were significantly increased(P<0.05),the changes of cell morphology and cytoskeleton remodeling were alleviated,P-cadherin protein expression were significantly increased(P<0.05),FSP-1 protein expression were significantly decreased(P<0.05).Conclusion:Drug-containing serum of TSN significantly inhibited podocyte EMT in high-glucose environment.
