摘 要:目的:研究蚕沙对湿阻中焦证大鼠肠道菌群的影响,探讨蚕沙和胃的作用机制,从肠道菌群的角度揭示粪便类中药的科学内涵。方法:应用PCR-DGGE技术获得肠道菌群分子指纹图谱,计算多样性指数、物种丰度、均匀度指数,并进行聚类分析;比较各组大鼠、蚕沙药材中的菌群分子指纹图谱的相似性。结果:与正常组比较,模型组大鼠多样性指数、物种丰度、均匀度指数均显著减少(P<0.05);与模型组比较,蚕沙中、高剂量组大鼠多样性指数、均匀度指数显著增加(P<0.05),且高剂量组大鼠的物种丰度也显著增加(P<0.05)。聚类分析显示,蚕沙高剂量组与正常组聚集成一簇,蚕沙低、中剂量聚集成一簇,模型组单独聚成一簇。菌群相似性显示,与正常组比较,模型组相似性系数显著降低(P<0.05);与模型组比较,蚕沙低、中、高剂量的相似性系数均显著升高(P<0.05);而蚕沙药材中菌群的相似性系数与蚕沙低、中、高剂量的相似性系数比较,差异均无统计学意义。结论:蚕沙可能是通过平衡肠道菌群而发挥和胃的作用,而这种作用可能与蚕沙中含有的菌群有关。Objective:To study the effects of Faeces Bombycis(FB)on the intestinal microflora of rats with syndrome of damp retention in middle-jiao,and to explore the mechanisms of‘harmonize the stomach’,and further reveal the scientific connotation of fecal Chinese medicine from the perspective of intestinal microflora.Methods:The molecular fingerprint of intestinal microflora was obtained by the PCR-DGGE technique.The Shannon-Wiener diversity,genotypic richness and Pielou evenness index were calculated,and the similarity of the intestinal microflora was analyzed by clustering;The similarity of the microflora between each group of rats and the FB was compared.Results:Compared with the control group,the Shannon-Wiener diversity index,genotypic richness and Pielou evenness index of the rats in the model group were significantly decreased(P<0.05);Compared with the model group,the Shannon-Wiener diversity index and Pielou evenness index of the middle and high dose groups of FB were significantly increased(P<0.05),and the genotypic richness of the high doses group was also significantly increased(P<0.05).Cluster analysis showed that the bacterial communities in high doses group of FB and the control group clustered closely to one another,and the bacterial communities in the low and medium doses group of FB were clustered together,while the bacterial communities in the model group was clustered into a category.The microflora similarity showed that compared with the control group,the similarity coefficient of the model group was significantly decreased(P<0.05),and compared with the model group,the similarity coefficient of the low,medium and high dose groups of FB were significantly increased(P<0.05);while the similarity coefficient of the microflora in FB were no apparent differences compared with the low,medium and high dose groups of FB.Conclusion:FB may play role on‘harmonize the stomach’by balancing intestinal microflora,which may be related to the microflora contained in the FB.
