摘 要:目的:探究南葶苈子药材中槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷(槲皮素双糖苷)的稳定性,为南葶苈子在临床上的合理应用提供借鉴和参考。方法:采用经典恒温法考察了该成分在水溶液中的热稳定性,得出了不同温度下的反应速率方程,推算出了其有效期和半衰期;以lgK(K为反应速率常数)对1/T进行一元线性回归,得到回归方程;考察该成分在不同pH、不同溶剂以及不同浓度条件下的稳定性情况。结果:该成分在80、100℃条件下加热10h后,含量分别降至初始含量的50.1%、5.4%,热降解规律符合零级反应方程。温度为60、80、100℃时的有效期分别为7.71、2.95、2.39h,半衰期分别为58.29、7.54、5.07h。另外,在pH>6条件下,该成分发生大量降解,为碱催化反应。结论:南葶苈子中的槲皮素双糖苷不稳定,在制剂生产过程中应进行相应条件控制。Objective:To explore the stability of quercetin-3-O-β-D-glucose-7-O-β-D-diglycoside,a medical material of Descurainia sophia seeds and provide reference for rational clinical application of Descurainia sophia seeds.Methods:Through the classical thermostatic method,we investigated the thermal stability of Quercetin-3-O-β-D-glucose-7-O-β-Ddiglycoside in aqueous solution,obtained the reaction rate equation at different temperature and calculated its validity and halflife time;Moreover,the linear regression of 1/T with lgK(K is the rate constant)and the regression equation was calculated;In addition,we investigated the stability of this ingredient under different pH values,solvents and concentrations.Results:After heating this component at 80℃and 100℃for 10 h,the content dropped to 50.08%and 5.4%of the original content,respectively,and the thermal degradation rule conformed to the zero-order reaction equation.When the temperature is 60,80,100℃,the validity period is 7.71,2.95,2.39 h,and the half-life is 58.29,7.54,5.07 h.In addition,under the condition of pH>6,a large amount of this component is degraded,which is an alkali-catalyzed reaction;Conclusion:Quercetin-3-O-β-Dglucose-7-O-β-D-gentiobioside in Descurainia sophia seeds is unstable,and corresponding conditions should be controlled during the preparation process.
