摘 要:目的:同时测定大黄、虎杖、决明子以及何首乌中蒽醌类物质的含量,并考察其体外抗氧化活性,分析蒽醌类成分的含量与体外抗氧化活性的关系。方法:采用HPLC-UV法测定4种药材中10种蒽醌类成分的含量;通过测定DPPH自由基清除率,评价4种药材提取物的体外抗氧化活性。结果:10种蒽醌在测定范围内线性关系较好(R2>0.999),精密度、重复性、稳定性及加样回收率均在要求范围内;蒽醌类成分种类:大黄>决明子>虎杖>何首乌;蒽醌苷总含量:虎杖>大黄>决明子>何首乌;游离蒽醌总含量:大黄>虎杖>决明子>何首乌。DPPH清除能力为:虎杖>大黄>决明子>何首乌;抗氧化活性与大黄素-8-O-β-D-葡萄糖苷及总蒽醌苷的含量呈显著正相关(P<0.01)。结论:蒽醌苷类成分可能是含蒽醌类成分中药的抗氧化活性成分,本研究为含蒽醌类成分药材的分析检测及天然抗氧化剂的开发提供了理论依据。Objective:To determine the contents of anthraquinones in Radix et Rhizoma Rhei,Rhizoma Polygoni Cuspidati,Semen Cassiae and Radix Polygoni Multiflori,and to investigate their antioxidant activities in vitro,and to analyze the relationship between the contents of anthraquinones and antioxidant capacity in vitro.Methods:The contents of 10 anthraquinones in four medicinal materials were determined by HPLC-UV.The DPPH radical scavenging rate was determined to evaluate the antioxidant activity of the extracts of four medicinal materials in vitro.Results:The linear relationship of 10 kinds of anthraquinones was good(R2>0.999),and the precision,repeatability,stability and recovery were all within the required range.The number of anthraquinone species was Radix et Rhizoma Rhei>Semen Cassiae>Rhizoma Polygoni Cuspidati>Radix Polygoni Multiflori;the total contents of anthraquinones was Rhizoma Polygoni Cuspidati>Radix et Rhizoma Rhei>Semen Cassiae>Radix Polygoni Multiflori;the total contents of free anthraquinones was Radix et Rhizoma Rhei>Rhizoma Polygoni Cuspidati>Semen Cassiae>Radix Polygoni Multiflori.The results showed that DPPH scavenging capacity was Rhizoma Polygoni Cuspidati>Radix et Rhizoma Rhei>Semen Cassiae>Radix Polygoni Multiflori.The correlation analysis showed that the antioxidant capacity was significantly correlated with the content of emodin-8-O-β-D-glucoside and total anthraquinone glycosides(P<0.01).Conclusion:Anthraquinone glycosides may be the antioxidant components of traditional Chinese medicine containing anthraquinones.This study provides a theoretical basis for the analysis and detection of anthraquinones and the development of natural antioxidants.
