摘  要:目的:研究南方红豆杉水提物联合紫杉醇对肺癌A549细胞增殖的抑制作用和对耐药相关基因的影响。方法:将A549细胞分为不同浓度南方红豆杉水提物组及空白对照组,用CCK-8法检测药物作用48h后对细胞存活率的影响。根据上述结果,再将A549细胞分为紫杉醇联合不同浓度南方红豆杉水提物组、紫杉醇组、空白对照组,用CCK-8法检测药物作用48h后对细胞存活率的影响,用RT-PCR检测细胞内ABCB1、ABCG2、ABCC1、Bcl-2、Bax基因表达变化。结果:紫杉醇10μg/mL(P)、P+红豆杉水提物400μg/mL、P+红豆杉水提物800μg/mL、P+红豆杉水提物1200μg/mL、P+红豆杉水提物1600μg/mL,各组对A549细胞的抑制率分别为:17.47%、53.09%、63.22%、76.56%、85.29%;不同浓度红豆杉联合紫杉醇引起ABCC1、ABCB1、Bcl-2基因的水平逐渐降低(P<0.01,P<0.05),呈剂量相关性,Bax基因的水平逐渐上升(P<0.01),呈剂量相关性,ABCG2基因的表达无明显变化。结论:南方红豆杉水提物联合紫杉醇能抑制A549细胞生长,并能下调ABCC1、ABCB1、Bcl-2基因的表达,上调Bax基因的表达。Objective:To study the effects of aqueous extract of Taxus chinensis.var(AETC)combining Paclitaxel on inhibitory action of proliferation of human lung carcinoma A549 cells and resistance genes.Methods:The A549 cells were divided into different concentrations of AETC groups and control group,detecting of drug effect of 48 h with the method of CCK-8 and the effect on cell survival.Based on the above results,then A549 cells were divided into Paclitaxel combining different concentrations of AETC groups,Paclitaxel group,control group,detecting of drug effect of 48 h with the method of CCK-8 and the effect on cells survival.The gene expression of ABCB1,ABCG2,ABCC1,Bcl-2,Bax were examined by RT-PCR.Results:Paclitaxel 10μg/mL(P),P+ATEC 400μg/mL,P+ATEC 800μg/mL,P+ATEC 1200μg/mL,P+ATEC 1600μg/mL,each group of A549 cell inhibition rate is respectively:17.47%,53.09%,63.22%,76.56%,85.29%;Different concentrations of Taxus chinensis.var combining Paclitaxel could regulate down the gene ABCC1,ABCB1,Bcl-2 expressions(P<0.01,P<0.05),regulate up the gene Bax expression(P<0.01),and correlated to the dose.Taxus chinensis.var combining Paclitaxel showed no effects on ABCG2 gene expression.Conclusion:Taxus chinensis.var combining Paclitaxel could inhibit the growth of A549 cells,and regulate down the resistance related gene ABCC1,ABCB1,Bcl-2 expressions,regulate up the gene Bax.
