摘 要:目的:通过观察哮喘模型大鼠体表敏化点的分布及敏化点处温度,结合敏化点处肥大细胞(MCs)和P物质(SP)变化特征,探讨哮喘模型大鼠穴位敏化点及热敏点的产生机制。方法:60只雄性SD大鼠随机均分6组,分别为模型Ⅰ组和空白Ⅰ组,模型Ⅱ组和空白Ⅱ组,模型Ⅲ组和空白Ⅲ组,每组10只。模型组予卵白蛋白/氢氧化铝混合液腹腔及四肢注射后超声雾化吸入卵白蛋白0.9%氯化钠溶液,制备哮喘大鼠模型。其中模型Ⅰ组连续造模4周,模型Ⅱ组连续造模8周,模型Ⅲ组连续造模12周;空白Ⅰ、Ⅱ、Ⅲ组分别注射溶液4周、8周、12周,注射方法同模型组。实验结束后沿大鼠尾静脉注射伊文思蓝(EB),观察大鼠皮肤EB渗出点,同时用红外热像仪观测EB渗出点处温度,并检测相应皮肤组织中MCs及SP的变化情况,分析腧穴敏化与热敏化的相关性。结果:模型组大鼠的肺俞穴、定喘穴以及华盖穴处出现敏化现象;与空白组相应点比较,模型组敏化点处温度显著升高(P<0.05),MCs数及其脱颗粒率显著增加(P<0.05),SP阳性目标平均光密度亦显著增加(P<0.05)。结论:哮喘模型大鼠的敏化穴有肺俞穴、定喘穴、华盖穴,热敏穴主要是肺俞穴和定喘穴,两者出现一定的重合;MCs和SP参与腧穴的敏化现象,且随着疾病程度的增加,腧穴的敏化表现一种扩散的现象。Objective:To explore the mechanism of acupoint sensitization and heat sensitization points in asthmatic rats by observing the distribution of sensitization points on the body surface of asthmatic rats and the temperature of the sensitization points,and to detect the change characteristics of mast cells(MCs)and substance P(SP)at the sensitization points.Methods:A total of 60 male SD rats were randomly divided into six groups,including model group I and control group I,model group II and control group II,model group III and control group III respectively(n=10).In the model group,ovalbumin/aluminum hydroxide mixed solution was injected into abdominal cavity and limbs and then ultrasonic atomization was adopted to inhale ovalbumin normal saline solution to prepare asthma rat model.Among them,model group I was continuously modeled for 4 weeks,model group II was continuously modeled for 8 weeks and model group III was continuously modeled for 12 weeks.Normal saline was injected into control group I,II and III for 4 weeks,8 weeks and 12 weeks respectively.The injection method was as the same as that of model group.After the experiment,Evans blue(EB)was injected along the tail vein of the rat to observe the EB exudation point of the rat skin.Then the infrared thermal imager was used to observe the temperature of EB exudation point,and the changes of MCs and SP in corresponding skin tissues were detected to analyze the correlation between acupoint sensitization and heat sensitization.Results:Sensitization appeared at the Feishu(BL 13),Dingchuan(EX-B1)and Huagai(CV 20)in the model group rats.Compared with the corresponding points in the control group,the temperature of the sensitization point in the model group increased significantly(P<0.05).The MCs number and its degranulation rate increased significantly(P<0.05).The average optical density of SP-positive targets also increased significantly(P<0.05).Conclusion:For asthma rats,sensitization acupoints include Feishu(BL 13),Dingchuan(EX-B1)and Huagai(CV 20).Heat sensitizati
