基于配伍环境的补中益气汤对洛哌丁胺大鼠便秘模型影响的方证相关性研究

摘　　要：目的:探讨不同剂量配伍环境下的补中益气汤对洛哌丁胺大鼠便秘模型的影响及现代医学内涵。方法:采用洛哌丁胺皮下注射法复制便秘大鼠模型,将模型动物随机分为模型组,高、中、低剂量(1.76、0.88、0.44g/d)组,大黄(12.5g/d)组,给药7d后处死并检测小肠推进率及血清胃动素(MTL)、胃泌素(GAS)、血管活性肠肽(VIP)的含量。结果:与正常组比较,模型组大鼠的小肠推进率及MTL、GAS显著下降(P<0.01),VIP显著升高(P<0.01)。与模型组比较,高、中、低组大鼠小肠推进率、血清MTL、GAS含量均呈升高趋势,血清VIP含量均呈下降趋势,且与剂量呈负相关,低剂量组效果显著(P<0.01);而大黄组大鼠小肠推进率降低(P<0.01),血清MTL、GAS含量显著降低(P<0.01,P<0.05)。结论:趋近原书剂量的补中益气汤对洛派丁胺大鼠便秘模型具有最佳的治疗作用,以方测证则该模型为脾虚证便秘模型。Objective:To investigate the effects of Buzhong Yiqi Decoction on the constipation model of loperamide rats with different doses synergy environment and the connotation of modern medicine.Methods:The rat model of constipation was replicated by subcutaneous injection of loperamide suspension.After successful modeling,the model animals were randomly divided into control group,model group,high,medium and low dose(1.76,0.88,0.44 g/d)group and rhubarb(12.5 g/d)group.After 7 days of administration,the rats were sacrificed,and the small intestinal propulsion rate,serum gastrointestinal hormones MTL,GAS,and VIP were measured.Results:Compared with the control group,the small intestinal propulsion rate,MTL and GAS of the model group were significantly decreased(P<0.01),and the VIP increased significantly(P<0.01).Compared with the model group,the small intestinal propulsion rate,MTL and GAS content of the rats in the Buzhong Yiqi Decoction group increased,the VIP content showed a downward trend,and was negatively correlated with the dose,and the effect was significant in the low-dose group(P<0.01);while in the rhubarb group,the small bowel advancing rate was reduced(P<0.01),and MTL and GAS content decreased significantly(P<0.01,P<0.05).Conclusion:Buzhong Yiqi Decoction with a dose approaching the original book has the best therapeutic effect on the constipation model of loperamide in rats,according to the theory of prescriptions and syndromes,the model is a constipation model with spleen deficiency syndrome.
