龟甲胶对胃溃疡模型小鼠治疗作用及机制研究

摘　　要：目的:探讨龟甲胶对胃溃疡模型小鼠的治疗作用及其机制。方法:以食用醋酸灌胃构建小鼠胃溃疡模型,实验设正常对照组、模型对照组、三九胃泰组、龟甲胶组,经灌胃给药或0.9%氯化钠溶液1周后,常规法检测体质量、脾/胸腺指数、乳糖酶活性、胃残留率、胃溃疡指数及胃溃疡抑制率,滴定法检测胃液酸度,HE染色法检测胃、小肠、结肠的病理变化,ELISA法检测血清白介素(IL)-1β、IL-6、IL-8和胃泌素的含量水平。结果:与正常对照组比较,模型对照组体质量、脾指数、胸腺指数显著下降(P<0.01),胃液酸度、胃溃疡指数、胃残留率、胃和小肠中乳糖酶活力、血清IL-1β、IL-8和胃泌素含量水平显著升高(P<0.05,P<0.01),胃组织黏膜充血水肿、黏膜上皮细胞丢失、炎症细胞浸润,小肠和结肠上皮结构紊乱,大量炎性细胞浸润。与模型对照组比较,龟甲胶组体质量、脾指数、胸腺指数显著升高(P<0.01),胃液酸度、胃溃疡指数、胃残留率、胃和小肠乳糖酶活力、血清IL-1β、IL-8和胃泌素含量水平显著降低(P<0.01,P<0.05),胃组织黏膜充血水肿减轻、炎性细胞浸润减少,小肠和结肠组织上皮结构完整,排列有序。结论:龟甲胶可通过降低胃液酸度,减少IL-1β、IL-8和胃泌素的分泌,降低溃疡指数,从而发挥对胃溃疡的治疗作用。Objective:To explore the therapeutic effect and mechanism of Testudinis Carapacis et Plastri Colla on gastric ulcer model mice.Methods:Gastric ulcer model mice was established by intragastric administration of acetic acid.The experiment consisted of normal control group,model control group,Sanjiu Weitai group and Testudinis Carapacis et Plastri Colla group.After intragastric administration or normal saline for 1 week,body weight,spleen index,thymus index,lactase activity,gastric residue rate,gastric ulcer index and gastric ulcer inhibition rate were measured by routine method.The acidity of gastric juice was detected by titration,the pathological changes of stomach,small intestine and colon were detected by HE staining,and the level of serum IL-1β,IL-6,IL-8 and gastrin were detected by ELISA.Results:Compared with the normal control group,the body weight,spleen index and thymus index in model control group significantly decreased(P<0.01).The total acidity of gastric acid,gastric ulcer index,gastric residual rate,lactase activity in stomach and small intestine,with the levels of serum IL-1β,IL-8 and stomach gastrin-releasing significantly increased(P<0.05,P<0.01).There were the gastric mucosa hyperemia and edema,loss of mucosal epithelial cells,inflammatory cells infiltration,small intestine and colon epithelium structure disorder,a large number of inflammatory cells in filtration.Compare with the model group,the body quality,spleen index and thymus index in Testudinis Carapacis et Plastri Colla group significantly increased(P<0.01).The total acidity of gastric acid,gastric ulcer index,gastric residual rate,lactase activity in stomach and small intestine,with the levels of serum IL-1β,IL-8 and stomach gastrin-releasing significantly decreased in Testudinis Carapacis et Plastri Colla group(P<0.01,P<0.05).The gastric mucosa hyperemia and edema were reduced,inflammatory cell infiltration was reduced,and the epithelial structure of small intestine and colon tissues was intact and orderly arranged.Conclusion:Testudin
