制附子中脂类生物碱快速提取及化学性质研究

摘　　要：目的:建立一种快速分离提取制附子中脂类生物碱的方法,研究脂类生物碱的化学性质。方法:首先用乙醇提取制附子中的总生物碱,再根据不同类型生物碱水溶性的差异将脂类生物碱与其它类型生物碱分离;利用电喷雾质谱方法研究脂类生物碱在高温、酸性、碱性条件下的稳定性及与棕榈酸的酯交换反应。结果:本实验建立了一种从附子中快速分离制备脂类生物碱的方法,没有单酯型生物碱和双酯型生物碱的干扰;脂类生物碱在高温和酸性条件下比较稳定,在碱性条件出现部分降解;脂类生物碱与棕榈酸等脂肪酸不会发生酯交换反应。结论:本实验方法可快速制备脂类生物碱,效率高,且制备的脂类生物碱具有能相对稳定存在的性质。实验结果为脂类生物碱在附子的炮制、配伍、药理作用、药用价值等方面的研究奠定了基础。Objective:A method for rapid separation and extraction of lipo-alkaloids from processed Aconiti Lateralis Radix Praeparata was established,the chemical properties of lipo-alkaloids were studied.Methods:Firstly,the alkaloids in processed Aconiti Lateralis Radix Praeparata were extracted with ethanol,and then the lipo-alkaloids were separated from other types of alkaloids according to the difference of water solubility of different types of alkaloids.The stability of lipoalkaloids under high temperature,acidic and basic conditions and the ester-exchange reactions with palmitic acid were studied by electrospray ionization mass spectrometry.Results:A method for the rapid separation and preparation of lipo-alkaloids from processed Aconiti Lateralis Radix Praeparata was established in this experiment.This method was highly efficient,and no interference from monoester alkaloids and diester alkaloids.It was found that lipo-alkaloids were stable under high temperature,acidic and basic conditions except that only a little of lipo-alkaloids discomposes under basic conditions and did not undergo esterexchange reactions with fatty acids such as palmitic acid.Conclusion:By the experimental method,lipo-alkaloids can rapidly be prepared without loss of lipo-alkaloids and residue of other alkaloids.The results indicate that lipo-alkaloids have relatively stable properties.The experimental results laid the foundation for the study of lipo-alkaloids in the compatibility of traditional Chinese medicine,pharmacological effects,medicinal value,and so on.
