摘 要:目的:研究黄芩-黄连药对不同配比2种主要降糖成分的含量差异及对HepG2细胞葡萄糖消耗量的影响,优选黄芩-黄连药对改善2型糖尿病的最佳比例并阐述其配伍规律。方法:采用液质联用法分析黄芩-黄连1∶1、黄芩-黄连1∶2与黄芩-黄连2∶1配比中小檗碱和黄芩苷含量变化;观测黄芩-黄连不同配比对HepG2细胞胰岛素抵抗模型葡萄糖消耗量的影响。结果:与黄芩-黄连1∶1比较,黄芩-黄连1∶2中黄芩苷含量下降,而小檗碱含量增加;黄芩-黄连2∶1中小檗碱和黄芩苷含量差异则较小。与模型组比较,黄芩-黄连1∶2、黄芩-黄连1∶1、黄芩-黄连2∶1高剂量组葡萄糖消耗量显著升高(P<0.01,P<0.05),且黄芩-黄连1∶1高剂量葡萄糖消耗量最多。结论:黄芩-黄连1∶1配比为降糖作用最佳配比,其可能配伍机制为黄芩苷对于小檗碱发挥降糖作用具有双向调节效应。Objective:To study the effects of different ratios of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma on the content of two major hypoglycemic components and the glucose consumption of HepG2 cells,and to optimize the best proportion of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma improving type 2 diabetes and explain its compatibility.Methods:The liquid chromatography-mass spectrometry was used to analyze the changes of berberine and baicalin content in the 1∶1,1∶2 and 2∶1 ratio of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma.And the effects of different ratios of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma on the glucose consumption of HepG2 cells in insulin resistance model were observed.Results:Compared with 1∶1 ratio of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma,the content of the two components in the couplet medicines of Scutellaria Radix and Coptidis Rhizoma 1∶2 was quite different,which the content of baicalin in the couplet medicines of Scutellaria Radix and Coptidis Rhizoma 1∶2 decreased,while the content of berberine increased.In addition,the difference in the content of berberine and baicalin between the couplet medicines of Scutellaria Radix and Coptidis Rhizoma 1∶1 and the couplet medicines of Scutellaria Radix and Coptidis Rhizoma 2∶1 was not obvious.Furthermore,1∶2,1∶1 and 2∶1 couplet medicines of Scutellaria Radix and Coptidis Rhizoma high-dose group had a significant difference in glucose consumption(P<0.01,P<0.05).Compared with the model group,the glucose consumption decreased most significantly in the couplet medicines of Scutellaria Radix and Coptidis Rhizoma 1∶1 high dose group.Conclusion:1∶1 is the best ratio of the couplet medicines of Scutellaria Radix and Coptidis Rhizoma for hypoglycemic effect,and the possible compatibility mechanism is that baicalin has a two-way regulation effect on the hypoglycemic effect of berberine.
