摘 要:目的:观察夹脊电针对肌萎缩侧索硬化(ALS)hSOD^(G93A)转基因小鼠腰髓星形胶质细胞升高基因-1(AEG-1)及PI3K/Akt信号通路的影响。方法:将36只SPF级hSOD1^(G93A)转基因阳性雄性小鼠随机分为模型组、针刺组、电针组,每组12只,同窝转基因阴性的野生型雄性小鼠12只为野生组。针刺组、电针组小鼠分别给予夹脊针刺和夹脊电针治疗,每周治疗2次,连续治疗4周,野生组、模型组不予针刺。于120日龄取小鼠脊髓腰段分别检测小鼠腰髓AEG-1表达和AEG-1及PI3K/Akt信号通路相关蛋白的表达。结果:与野生组比较,模型组小鼠腰髓组织AEG-1、p-Akt、p-CREB表达显著降低(P<0.01),p-GSK-3β表达显著升高(P<0.01);与模型组比较,针刺组、电针组小鼠腰髓组织AEG-1、p-Akt、p-CREB表达显著升高(P<0.05,P<0.01),p-GSK-3β表达显著降低(P<0.05,P<0.01),且电针组效果明显。结论:夹脊电针能够促进ALS-hSOD1^(G93A)转基因小鼠腰髓AEG-1的表达,调节PI3K/Akt信号通路相关蛋白的表达,发挥运动神经元保护作用。Objective:To observe the effects of Jiaji(EX-B2)electro-acupuncture on AEG-1 and PI3 K/Akt signaling pathway in lumbar spinal cord of ALS hSOD1G93A transgenic mice.Methods:Thirty-six SPF grade hSOD1G93A transgenic positive male mice were randomly divided into model group,acupuncture group,electro-acupuncture group,12 mice in each group,with 12 transgenic negative male mice as wild type group.Acupuncture group and electro-acupuncture group mice were given acupuncture and electro-acupuncture on Jiaji(EX-B2)points,twice a week,and continued treatment of 4 weeks,wild type group and model group didn’t receive acupuncture treatment.At the age of 120 days,the expression of AEG-1 and PI3 K/Akt signaling pathway proteins in the lumbar spinal cord was detected.Results:Compared with the wild type group,the expression of AEG-1,p-Akt and p-CREB in the lumbar spinal cord tissue of model group were significantly decreased(P<0.01),and the expression of p-GSK-313 was significantly increased(P<0.01).Compared with model group,the expression of AEG-1,p-Akt and p-CREB in the lumbar spinal cord tissue of acupuncture and electric acupuncture groups were significantly increased(P<0.05,P<0.01),the expression of p-GSK-3βwas significantly decreased(P<0.05,P<0.01),and there was more significant effect in electric acupuncture group.Conclusion:Jiaji(EX-B2)electro-acupuncture can promote the expression of AEG-1 in lumbar spinal cord of ALS-hSOD1G93 A transgenic mice,regulate the expression of PI3 K/Akt signaling pathway related proteins,and play a protective role in motor neurons.
