摘  要:目的建立中药体外抑菌实验的初筛方法,并利用该方法从24味目前已知有抑菌、杀菌作用的中药中筛选出对碳青霉烯类药物耐药的肺炎克雷伯菌体外有抑制作用的中药。方法采用纸片扩散法进行初筛实验,并用微量肉汤稀释法对筛选出的对碳青霉烯类药物耐药的肺炎克雷伯菌体外有抑制作用的中药测定最低抑菌浓度。结果从24味中药中共筛选出黄连、连翘、紫花地丁3味对碳青霉烯耐药的肺炎克雷伯菌体外有抑制作用的中药,其余药物在体外对碳青霉烯耐药的肺炎克雷伯菌没有抑制作用。其中黄连对菌株9322和菌株9513的最小抑菌浓度均为0.0625 g/ml,连翘对菌株9322的最小抑菌浓度为0.25 g/ml、对菌株9513的最小抑菌浓度为0.0625 g/ml,紫花地丁对菌株9322和菌株9513的最小抑菌浓度均为0.0625 g/ml。结论纸片扩散法实验作为中药体外抑菌的初筛实验,可从大量的中药中筛选出对该菌体外具有抑制作用的中药。且操作简便、结果简单明了、重复性强,可作为中药筛选的平台。黄连、连翘、紫花地丁等中药对碳青霉烯耐药的肺炎克雷伯菌体外有一定的抑菌作用,为临床治疗对碳青霉烯耐药的肺炎克雷伯菌感染提供了参考方法。Objective To establish a primary screening method for the bacteriostatic experiment of traditional Chinese medicine in vitro,screen 24 kinds of traditional Chinese medicines having antibacterial and bactericidal effects with this method in order to find out the medicines that have bacteriostatic effects on Carbapenem-resistant Klebsiella pneumoniae in vitro.Methods The initial diffusion experiment was carried out by the paper diffusion method,and the minimum inhibitory concentration of the selected Chinese medicines that have bacteriostatic effects on the Carbapenem-resistant Klebsiella pneumoniae in vitro was determined by the micro-broth dilution method.Results Three kinds of Chinese medicines having bacteriostatic effects on Carbapenem-resistant Klebsiella pneumoniae in vitro were selected from 24 kinds of Chinese medicines,including Coptis chinensis,Forsythia suspensa,and Viola fruticosa.Other medicines had no bacteriostatic effects on Carbapenem-resistant Klebsiella pneumoniae in vitro.The minimum inhibitory concentrations of Coptis chinensis in strain 9322 and 9513 were 0.0625 g/ml.The minimum inhibitory concentrations of Forsythia suspensa in strain 9322 and 9513 were 0.25 g/ml and 0.0625 g/ml,respectively.The minimum inhibitory concentrations of Viola fruticosa in strain 9322 and 9513 were 0.0625 g/ml.Conclusion The paper diffusion experiment,a preliminary screening experiment of traditional Chinese medicine bacteriostasis in vitro,screened a large number of traditional Chinese medicines that have bacteriostatic effects on Carbapenem-resistant Klebsiella pneumoniae.Due to its simplicity of operation,concise and explicit results and high repeatability,the method can be used in the screening of Chinese medicines.Coptis chinensis,Forsythia suspensa,and Viola fruticosa have certain bacteriostatic effects on Carbapenem-resistant Klebsiella pneumoniae in vitro,which provides a reference for the clinical treatment of infection of Carbapenem-resistant Klebsiella pneumoniae.
