摘 要:通过qPCR技术检测NPRB和NPRC在矮小品系(S3系)和隐性白羽肉鸡(RR)胸肌组织中的表达模式,结合C-型利钠肽(CNP)激素对成肌细胞分化,脂滴沉积和分解的作用,阐明CNP对肉鸡肌肉发育的调控作用。结果显示:在19日胚龄、4.8和16周,NPRB和NPRC mRNA在S3系鸡胸肌组织中的表达显著高于RR鸡(P<0.05);CNP外源诱导鸡的原代成肌细胞6 d后,与对照组相较,成肌细胞分化标志基因(MYOG和MYOD)的表达显著升高(P<0.01),但成肌细胞的脂滴沉积能力显著降低(P<0.05)、培养基中甘油浓度显著升高(P<0.05)。以上结果提示,CNP通过促进成肌细胞的分化,抑制脂滴沉积,促进脂分解的作用,进而调控骨骼肌的发育。In this study, the expression pattern of NPRB(CNP receptor) and NPRC(clearance receptor) in the pectinal tissues was detected in S3(sex-linked dwarf chicken) and RR(recessive White Rock chickens) chickens using qPCR. Combined with the effects of CNP on myoblast differentiation, lipid droplet deposition and decomposition, the regulatory function CNP on skeletal muscle development was elucidated. The results showed that the expression of NPRB and NPRC mRNA in the breast muscle of S3 strain chickens was significantly higher than that of RR chickens(P<0.05) at developmental stages of E19, 4 W, 8 W and 16 W;Expression of MyoG and MyoD(marker genes for myoblasts differentiation) was significantly higher than that of the control group(P<0.01) after 6 days of CNP induction. However,the ability of lipid droplet deposition of myoblasts decreased significantly(P<0.05), and the glycerolconcentration in the medium increased significantly(P<0.05). These results suggested that CNP regulates the development of skeletal muscle through up-regulating the differentiation of myoblasts, inhibiting the lipid drop deposition of myoblasts and promoting lipid decomposition.
