摘  要:目的探讨黄芪葛根配伍对糖尿病大鼠脂肪组织胰岛素信号通路的影响,明确其对血糖调控机制。方法将86只大鼠随机分为6组,除正常组11只外,其余各组15只大鼠予一次性腹腔注射2%STZ(46 mg/kg)造模。同日正常组、模型组予蒸馏水蒸馏水10 mL/kg,各给药组分别予金芪降糖片混悬液1.47 g/kg,黄芪2.7 g/kg,葛根1.35 g/kg,黄芪葛根汤4.05 g/kg,给药30 d。酶联免疫吸附测定(ELISA)检测脂肪组织中胰岛素(Ins)、Ins受体(InsR)、葡萄糖转运体4(Glut-4)的水平,实时荧光定量聚合酶链式反应法(Real-time PCR)检测脂肪组织中Ins底物(IRS)、磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(PKB/AKT)mRNA相对表达。结果与正常大鼠比较,模型大鼠Ins、InsR、Glut-4水平降低,IRS、PI3K、AKT mRNA表达减少(P<0.05)。与模型大鼠比较,黄芪、葛根及其配伍能够升高糖尿病大鼠Ins(葛根、黄芪与葛根配伍除外)、InsR、Glut-4(葛根除外)水平(P<0.05),在调节InsR、Glut-4时,黄芪优于两药合用;黄芪、葛根及两药配伍能够增加IRS(葛根、黄芪与葛根配伍除外)、PI3K(葛根除外)、AKT mRNA表达(P<0.05),黄芪促进PI3K mRNA表达的作用优于两药合用,黄芪与葛根配伍调节AKT mRNA表达的作用优于单一用药。结论黄芪、黄芪与葛根配伍能够调节糖尿病大鼠脂肪组织胰岛素信号通路,黄芪作用效果佳。Objective To observe the effect of Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata) compatibility on insulin signaling pathway in adipose tissue of diabetic rats and discuss its mechanism of reducing blood glucose. Methods Eighty-six rats were randomly divided into six groups. There were 11 rats in normal group and 15 rats in the other groups. Except for the normal group, 2% STZ(46 mg/kg) was given to rats of the other groups by intraperitoneal injection to induced diabetes models. On the same time, 10 mL/kg distilled water was given to rats in the normal group and the model group were given 1.47 g/kg suspension of Jinqi Jiangtang Tablet. Huangqi(Radix Astragali) with the dose of 2.7 g/kg was given to rats in Huangqi(Radix Astragali) group. Gegen(Radix Pueraria Lobata) with th dose of 1.35 g/kg was given to Gegen(Radix Pueraria Lobata) group. Huangqi Gegen Decoction with the dose of 4.05 g/kg was given to Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata) compatibility group. All of drugs were given for 30 days continuously. The contents of insulin(Ins), insulin receptors(InsR) and glucose transporter4(Glut-4) were detected by enzyme-linked immunosorbent assay(ELISA). The relative expression of substrate of InsR(IRS), phosphatidylinositol 3 kinase(PI3 K) and protein kinase B(PKB/AKT) mRNA were tested by Real-time PCR. Results Compared with those of the normal rats, the contents of Ins, InsR and Glut-4 were lowered, while the relative expressions of IRS, PI3 K and AKT mRNA were reduced of model rats(P<0.05). Huangqi(Radix Astragali),Gegen(Radix Pueraria Lobata) and their compatibility could increase Ins[except Gegen(Radix Pueraria Lobata) and Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata) compatibility]. InsR and Glut-4[except Gegen(Radix Pueraria Lobata)] of model rats(P<0.05). Huangqi(Radix Astragali) regulated InsR and Glut-4 better than Huangqi(Radix Astragali)-Gegen(Radix Pueraria Lobata) compatibility.
