摘 要:目的观察胃痛消痞方对肝郁脾虚型功能性消化不良(functional dyspepsia,FD)大鼠五羟色胺(5-HT)及其受体(5-HT3aR)表达的影响。方法选用改良郭氏夹尾刺激法联合张氏不规则喂食法复制FD大鼠模型,以西药莫沙必利片为阳性对照,中药干预以胃痛消痞方灌胃治疗。实验结束后,观察各组大鼠的一般状态,免疫组化法(IHC)观察下丘脑及胃窦组织5-HT蛋白表达,聚合酶链反应法(PCR)观察下丘脑及胃窦组织中5-HT3aR mRNA的变化。结果模型空白组大鼠一般状态差,下丘脑及胃窦组织中5-HT蛋白表达均下降,下丘脑及胃窦组织中5-HT3aR mRNA表达均降低(P<0.05);胃痛消痞方组及莫沙必利组大鼠一般状态明显改善,下丘脑及胃窦组织中5-HT蛋白表达均升高,下丘脑及胃窦组织中5-HT3aR mRNA表达均升高(P<0.05)。结论胃痛消痞方能上调FD大鼠5-HT及其受体5-HT3aR mRNA的表达,可能是其治疗FD的作用机制之一。Objective To observe the effects of Weitong Xiaopi Prescription on the expressions of 5-HT and receptor in the rats with liver-Qi depression and spleen deficiency type of functional dyspepsia(FD). Method GUO’s clip tail stimulation combined with ZHANG’s irregular feeding method was used to create rat model of liver-Qi depression and spleen deficiency. Mosapride was a positive control drug and Weitong Xiaopi Prescription was used to treat FD. We observed the general conditions of rats in all groups and the protein expressions of 5-HT in the hypothalamus and gastric antrum were detected by IHC and the expressions of 5-HT3 aR mRNA in the hypothalamus and gastric antrum were detected by PCR. Result The general conditions of model group was bad. The protein expressions of 5-HT in the hypothalamus and gastric antrum were decreased and the expressions of 5-HT3 aR mRNA in the hypothalamus and gastric antrum were decreased(P<0.05). The general conditions of Weitong Xiaopi Prescription group and mosapride group were improved. The protein expressions of 5-HT in the hypothalamus and gastric antrum were increased and expressions of 5-HT3 aR mRNA in the hypothalamus and gastric antrum were increased(P<0.05). Conclusion Weitong Xiaopi Prescription can increase the expressions of 5-HT and its receptor 5-HT3 aR mRNA in FD rats,which may be one of the mechanisms of treating FD.
