摘 要:为了探讨不同种属布鲁氏杆菌宿主特异性的分子遗传基础以及获得分子生物学种属鉴别的靶位点,试验采用相关生物信息学软件对GenBank中牛布鲁氏杆菌2308(B.abortus 2308)和羊布鲁氏杆菌16M(B.melitensis 16M)进行了全基因组范围内的单核苷酸多态性(SNPs)、插入缺失长度多态性(InDels)以及获得与缺失多态性(PAVs)分析。结果表明:B.abortus 2308和B.melitensis 16M之间存在SNPs 6 271个,InDels 2 799个;B.abortus 2308和B.melitensis 16M之间含有32个PAVs片段(大于100 bp),长度共计约57.5 kb。随机选取5个PAVs片段进行PCR扩增和测序,核酸序列结果与生物信息学分析结果一致,扩增长度与生物信息学分析结果之间的差异与PAVs片段两端存在的正向重复序列有关。说明本试验获得了B.abortus 2308和B.melitensis 16M全基因组范围内的SNPs、InDels以及PAVs,以利于下一步解析牛和羊布鲁氏杆菌种属生物学差异的遗传基础以及建立种属鉴别方法。In order to investigate the molecular genetic basis of host specificity of different Brucella species and to obtain the target sites for molecular biological identification among different species,in this study the single nucleotide polymorphisms(SNPs),insertion-deletion polymorphisms(InDels)and presence/absence variants(PAVs)between B.abortus 2308 and B.melitensis 16M from GenBank were analyzed at the bioinformatics software.The results showed that a total of 6271 SNPs and 2799 InDels were identified between B.abortus 2308 and B.melitensis 16M,respectively.Thirty-two PAVs(over 100 bp)with a total length of about 57.5 kb were identified between B.abortus 2308 and B.melitensis 16M.Five PAVs fragments were randomly selected for PCR amplification and sequencing.The nucleic acid sequence results were consistent with the bioinformatics analysis results,and the difference in amplification length and bioinformatics analysis results was related to the forward repeat sequence at both ends of PAVs fragment.These results revealed that the genome-wide range of SNPs,InDels and PAVs of B.abortus 2308 and B.melitensis 16M were obtained,which provided a theoretical basis for the further analysis of the biological genetic differences and the establishment of species identification method between B.abortus and B.melitensis.
