摘　　要：犬1型腺病毒E1区包括E1A、E1B-55K和E1B-19K。为了研究E1B-55K可延缓细胞过早凋亡,并协助mRNA翻译的作用。本文采用生物信息学方法预测E1B-55K蛋白的理化性质、亲疏水性、跨膜区结构、二级及三级结构、磷酸化及糖基化位点和抗原表位,并分析其结构和功能。结果发现E1B-55K有424个氨基酸,原子组成为C2199H3535N575O538S39,分子量为47887.13,带负电与正电的氨基酸残基总数分别为25和46个,不稳定指数为42.53。E1B-55K蛋白有23个磷酸化位点。只有1个糖基化位点位于第64位。E1B-55K是平均亲水系数为0.662的疏水蛋白,无信号肽和跨膜区结构。二级结构中折叠占12.50%、螺旋占38.68%、无规卷曲和延伸链分别为23.58%和25.24%。在三级结构中获得E1B-55K蛋白的可信度和覆盖率分别为38.6%和2%。有17个抗原决定簇和45个优势的B细胞抗原表位存在于E1B-55K蛋白中。从而得出预测E1B-55K蛋白的性质、结构和功能有助于建立基于E1B-55K的ELISA检测方法和亚单位疫苗的结论。Canine adenovirus 1E1 region including E1A,E1B-55K and E1B-19K.In order to study the role of the E1 B-55 K such as it can prevent premature cell apoptosis and participate in the translation of m RNA.Bioinformatics methods were used to predict the character of the E1 B-55 Kprotein,such as the physical and chemical properties,hydrophobicity,transmembrane region structure,secondary and tertiary structure,phosphorylation and glycosylation sites,and antigen epitopes,and analyze its function and structure.The results showed that E1 B-55 K had 424 amino acids.The atomic composition was C2199H3535N575O538S39,the molecular weight was 47887.13,the number of negatively and positively charged amino acids were 25 and 46,and the instability index was 42.53.E1B-55 K had 23 phosphorylation sites.There was a potential glycosylation site at position 64.E1B-55K is a hydrophobic protein with an average hydrophilic coefficient of 0.662,without signal peptide and transmembrane structure.In the secondary structure,the Beta-folding accounted for 12.50%,the Alpha helix accounted for 38.68%,the Random curl and the extended chain accounted for 23.58%and 25.24%,respectively.The reliability and coverage of E1B-55K protein were 38.6%and 2%,respectively.After antigen prediction,it was found that E1B-55K protein had 17 antigen determination clusters and 45 dominant B cell epitopes.It is concluded that to obtain the character,structure and function are helpful to build the ELISA method and subunit vaccine for CAdV-1.
