摘 要:目的观察microRNA-105-3p(miR-105-3p)在多囊卵巢综合征(PCOS)患者外周血中的表达特征及其对体外人卵巢颗粒细胞(KGN)增殖和凋亡的影响。方法选择PCOS患者作为PCOS组,同龄健康女性作为对照组,记录临床信息和生化指标,并采用荧光定量PCR(qRT-PCR)分析两组中miR-105-3p的表达量。再将miR-105-3pmimics转染KGN细胞,采用MTT比色法和酶联免疫吸附测定观察miR-105-3p表达改变对KGN细胞增殖和凋亡的影响。结果与对照组比较PCOS组患者体重指数、黄体生成素、雌二醇、催乳素、总睾酮、游离睾酮含量、卵巢体积和卵巢滤泡数均较高,而卵泡刺激素含量和每年月经数较低(P<0.05)。miR-105-3p在PCOS组中的表达量显著低于对照组(P<0.05)。KGN细胞转染后,miR-105-3p mimics中miR-105-3p的表达量显著高于mimics阴性对照(P<0.05)。MTT比色法显示体外转染miR-105-3p mimics至KGN细胞可显著抑制细胞增殖(P<0.05)。miR-105-3p mimics转染的KGN细胞中Caspase-3、Bax显著高于mimics阴性对照(P<0.05),Bel-2反之。结论miR-105-3p在PCOS外周血中表达下调,体外过表达miR-105-3p可显著抑制KGN细胞生长并促进凋亡,捉示miR-105-3p与PCOS病理过程密切相关,并可能作为PCOS诊治的新候选物标志物。Objctive To detect the exresion of microRNA-105-3p(miR-105-3p)in peripheral blood of patients with polycystic ovary syndrome(PCOS)。and to observe the efects on the proliferation and apoptosis of human ovarian granulosa cells(KGN)in vitro.Methods PCOS patients who came to our hospita's reproductive center were cllcted as the PCOS group.Healthy women of the same age was used as the control group.The clinical information and biochemical data were recorded in detail.Expression of miR-105-3p was amalyzed by real-time PCR(qRT-PCR).KGN cells were tansfected with miR-105-3p mimics.The efects of miR-105-3p on the proliferation and apoptosis of KGN cells were observed by MTT colorimetric and enzyme-linked immunosorbent assays.Results Compared with the control group,the PCOS group had higher body mass index,luteotropic hormone,estradiol,prolactin,total testosterone,free testosterone,ovarian volume and ovarian ollicular number,while fllicle stimulating hormone and number of menses per year were lower(P<0.05).The expression level of miR-105-3p in the PCOS group were lower than that in the control group(P<0.05).After KGN cell transfection,the expression level of miR-105-3p mimics was significantly higher than that in mimics negative control(P<0.05).In vitro transfection of miR-105-3p mimics to KGN cells signific antly inhibited cell growth compared to the mimics negative control treated cells(P<0.05).In miR-105-3p mimics transfected KGN cll,Caspase-3 and Bax were signifcantly higher than mimics negative control(P<0.05),and Bc-2 were lower.Conclusion The expression of miR-105-3p is down-regulated in the peripheral blood of PCOS patients.Overexpression of miR-105-3p in vitro signifcandly inhibits the prolifeaion of KGN cells and promotes apoptosis,suggsing that miR-105-3p is closely related to the pathological process of PCOS and may be used as a new candidate biomarker for diagnosis and treatment of PCOS.
