摘  要:目的研究白芍总苷(TGP)预处理对缺血性脑损伤大鼠脑组织的保护作用及其机制。方法240只SD大鼠随机分为假手术组、模型组、尼莫地平(NMP)10mg/(kg·d)组和TGP 50、100、200mg/(kg·d)组,每组40只;采用线栓阻断大脑右侧中动脉的方法制备缺血性脑损伤大鼠模型,造模手术前7天开始1次/天灌胃给药。24h后,Zausinger六分法评估神经功能,伊文思蓝渗透法测定血-脑脊液屏障(BBB)通透性,干湿比重法测定脑组织含水量,HE染色法行脑组织病理学检查,TUNEL法观察神经细胞凋亡状况,生化分析法测定脑组织抗氧化酶活性和MDA含量。结果与模型组比较,TGP 100、200mg/(kg·d)和NMP 10 mg/(kg·d)组大鼠Zausinger评分升高,伊文思蓝含量和含水量降低,脑组织病理性改变和神经细胞凋亡状况明显改善,凋亡指数(AI)降低,SOD、CAT活性升高且MDA含量降低,差异有统计学意义(P<0.05)。与NMP 10mg/(kg·d)组比较,TGP 200mg/(kg·d)组Zausinger评分升高,伊文思蓝含量降低,脑组织病理性改变明显改善,AI降低,SOD活性升高且MDA含量降低,差异有统计学意义(P<0.05)。结论TGP对缺血性脑损伤大鼠脑组织具有保护作用,其机制可能与抑制氧化应激、保护BBB通透性、抑制脑水肿有关。Objective To study the protective effects of Total Glucosides of Paeony(TGP)on brain tissue of the rats with ischemic brain injury and its possible mechanism.MethodsTotally 240 experimental SD rats were randomly divided into sham operation group,model group,nimodipine(NMP)10mg/(kg·d)and TGP 50,100,200mg/(kg·d)group(n=40).The rat model with ischemic brain injury was established by blocking the right middle cerebral artery,and the drugs were given by intragastric administration for 7d before modeling.The evaluation of neurological was assessed by Zausinger score.The permeability of BBB was measured by Evans blue permeation method and the water content of brain tissue was measured by dry wet specific gravity method.The pathological change of brain tissue was examined by HE staining and the apoptosis was observed by TUNEL method.The antioxidant enzyme activity and MDA content in brain tissue were determined by biochemical analysis.ResultsCompared with model group,the Zausinger score was increased and permeability of Evans blue,water content were decreased of the rats in TGP 100,200mg/(kg·d)and NMP 10mg/(kg·d)group.The pathological changes and neuron apoptosis was improved,and the AI was decreased.The activity of SOD,CAT were increased and the content of MDA was decreased.All of the differences were significant(P<0.05).Compared with NMP 10mg/(kg·d)group,the Zausinger score was increased and permeability of Evans blue was decreased of the rats in TGP 200mg/(kg·d)group.The pathological changes was improved,and the AI was decreased.The activity of SOD was increased and the content of MDA was decreased.All of the differences were significant(P<0.05).ConclusionTGP has protective effect on brain tissue of the rats with ischemic brain injury.The mechanism may be related to the inhibition of oxidative stress,protection of BBB permeability and the inhibition of cerebral edema.
