摘 要:用高效液相色谱法(HPLC)对青霉素V钾的发酵液及提取过程中相关物质进行了跟踪检测,找到了青霉素V钾及相关物质的变化规律,研究了青霉素V钾成盐溶液终点pH值、成盐溶液浓度、加碱时间对料液中任意杂质的影响,最佳工艺条件为青霉素V钾成盐溶液终点pH值6.5~6.8,成盐溶液浓度控制在300000 U/mL以内,加碱时间控制在40 min左右可以有效控制杂质1的含量,减少青霉素V钾的降解。The fermentation liquid penicillin V potassium and related substance of in the process of extraction was determined by high performance liquid chromatography(HPLC).The changing of profiles penicillin V and related substances in the process was elucidated.The influence of solution pH,solution concentration,alkali adding time on any impurities in the material liquid was studied.The optimal process conditions were as follows:solution pH was 6.5~6.8,solution concentration was within 300000 U/mL,the alkali adding time was within 40 min.The content of impurity 1 could be effectively controlled and the degradation of penicillin V potassium was reduced.
