摘  要:目的探讨柚皮苷(NAR)对妊娠期糖尿病小鼠胰岛素抵抗(IR)的改善作用机制。方法按照体重将受孕后的C57小鼠随机分为3组:正常组、模型组和实验组,每组17只。实验组灌胃给予NAR 50 mg·kg-1,正常组和模型组灌胃给予等量0.9%NaCl。3组小鼠均干预14 d。以血糖仪测定空腹血糖(FPG),以全自动电化学发光分析法测定空腹胰岛素(Fins),计算胰岛素抵抗(HOMA-IR)指数,以蛋白质印迹法检测骨骼肌腺苷酸蛋白激酶(AMPK)和葡萄糖运载体4(GLUT4)蛋白表达水平。结果干预14 d,正常组、模型组和实验组的FPG水平分别为(5.40±0.48),(16.18±2.37)和(9.86±2.32)mmol·L^-1;上述3组而的HOMA-IR指数分别为2.11±0.52,7.11±0.93和3.97±0.85;上述3组骨骼肌p-AMPK/AMPK蛋白比值分别为0.84±0.09,0.34±0.07和0.61±0.07;上述3组GLUT4蛋白表达水平分别为0.92±0.11,0.56±0.10和0.75±0.10。上述指标:模型组与正常组比较,差异均有统计学意义(均P<0.05);实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论柚皮苷可改善妊娠期糖尿病小鼠模型体重和血糖水平,减轻IR,激活AMPK-GLUT4途径、促进葡萄糖代谢可能是降低IR的重要作用机制。Objective To investigate the effect and mechanism of naringin(NAR) on insulin resistance(IR) in gestational diabetes mellitus mice. Methods C57 mice after conception were randomly divided into three groups according to weight:normal group, model group and experimental group,17 mice in each group. The experimental group was given NAR(50 mg·kg-1) by gavage, the normal group and model group were given the same amount of saline by gavage,for 14 d. The fasting plasma glucose(FPG) was measured by blood glucose meter. Fasting insulin(Fins) was measured by full-automatic electrochemiluminescence analysis. Index of the insulin resistance(HOMA-IR) was calculated. The protein expression of AMP-activated protein kinase(AMPK) and glucosetransporter 4(GLUT4) in skeletal muscle was detected by Western blotting. Results On day 14, FPG levels in normal group, model group and experimental group were(5.40±0.48),(16.18±2.37) and(9.86±2.32) mmol·L^-1,respectively;index of the HOMA-IR in the three groups were 2. 11 ± 0. 52,7. 11 ± 0. 93 and 3. 97 ± 0. 85,respectively;p-AMPK/AMPK protein ratio in the three groups were 0. 84 ± 0. 09,0. 34 ± 0. 07 and 0. 61 ± 0. 07,respectively;expression levels of GLUT4 in the three groups were 0. 92 ± 0. 11,0. 56 ± 0. 10 and 0. 75 ± 0. 09,respectively. Comparison between normal group and model group,the difference of the factors were significant(all P < 0. 05);comparison between experimental group and model group,the difference of the factors were significant(all P < 0. 05). Conclusion The naringin can improve body mass and blood glucose level of gestational diabetes mellitus mice,reduce IR,activate AMPK-GLUT4 pathway and promote glucose metabolism,which may be an important mechanism of IR reduction.
