摘 要:目的研究枳实总黄酮苷对胆汁反流性胃炎(BRG)大鼠的胃黏膜保护作用及增殖细胞核抗原(PCNA)、C-erbB-2表达的影响。方法用反流液诱导构建BRG大鼠模型,将成功建立的BRG大鼠随机分为模型组与实验组,各20只,另选20只未加任何干预的大鼠作为对照组。实验组大鼠灌胃150 mg·kg-1·d-1枳实总黄酮苷,对照组与模型组灌胃等量生理盐水,每天1次,连续干预14 d。用酶联免疫吸附(ELISA)法检测胃黏膜组织前列腺素E2(PG E2)、细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)与白细胞介素-8(IL-8)含量;用免疫组化法检测胃黏膜组织PCNA、C-erbB-2蛋白的表达情况;用苏木精-伊红(HE)染色观察大鼠胃黏膜组织病理学变化。结果对照组、模型组与实验组大鼠胃黏膜组织中PGE2含量分别为(194.22±21.37),(79.62±8.53),(126.65±9.84)pg·mg-1;ICAM-1含量分别为(19.11±2.13),(46.83±5.18),(30.02±2.59)pg·mg-1;TNF-α含量分别为(9.32±2.69),(36.74±5.45),(21.33±3.21)pg·mg-1;IL-8含量分别为(5.89±1.13),(26.72±4.27),(14.12±1.85)pg·mg-1;胃黏膜组织PCNA蛋白阳性细胞百分比分别为(6.21±2.02)%,(39.43±3.11)%,(18.73±3.06)%;C-erbB-2蛋白阳性细胞百分比分别为(10.22±2.16)%,(51.86±4.05)%,(26.31±3.39)%。与对照组比较,模型组大鼠胃黏膜组织中ICAM-1、TNF-α、IL-8含量及PCNA、C-erbB-2蛋白阳性细胞百分比均显著升高,而实验组低于模型组;模型组大鼠胃黏膜组织PGE2含量低于对照组,而实验组高于模型组(均P<0.05)。结论枳实总黄酮苷可显著抑制BRG大鼠胃黏膜组织炎性因子水平,减轻炎症反应,同时降低胃黏膜组织PCNA、C-erbB-2蛋白表达,从而发挥胃黏膜保护作用。Objective To investigate the protective effect of total flavonoid glycosides of Zhishi on gastric mucosa and expression of proliferating cell nuclear antigen(PCNA) and C-erbB-2 in rats with bile reflux gastritis(BRG). Methods The BRG rat models were constructed using reflux fluid induction, the successfully established BRG rats were randomly divided into model group(n=20) and test group(n=20), and another 20 rats without any intervention were selected as control group. Rats in test group were given with total flavonoid glycosides of Zhishi 150 mg·kg-1·d-1 by gavage, control group and model group were given the same volume of normal saline by gavage,1 time·d-1 and intervened continuously for 14 d. Enzyme-linked immunosorbent assay(ELISA) was used to detect prostaglandin E2(PGE2),intercellular adhesion molecule-1(ICAM-1),tumor necrosis factor-α(TNF-α) and interleukin-8(IL-8) content;the expression of PCNA and C-erbB-2 protein in gastric mucosa tissues were detected by immunohistochemistry method;hematoxylin-eosin(HE) staining was used to observe the histopathological changes of rat gastric mucosa tissues. Results The contents of PGE2 in the gastric mucosa tissues of control group,model group and test group were(194. 22 ± 21. 37),(79. 62 ± 8. 53),(126. 65 ± 9. 84) pg·mg-1;the contents of ICAM-1 were(19. 11 ± 2. 13),(46. 83 ± 5. 18),(30. 02 ± 2. 59) pg·mg-1;the content of TNF-α were(9. 32 ± 2. 69),(36. 74 ± 5. 45),(21. 33 ± 3. 21) pg·mg-1;the content of IL-8 were(5. 89 ± 1. 13),(26. 72 ± 4. 27),(14. 12 ± 1. 85) pg·mg-1;the percentage of PCNA protein positive cells in gastric mucosa tissues were(6. 21 ± 2. 02) %,(39. 43 ± 3. 11) %,(18. 73 ± 3. 06) %;the percentages of C-erbB-2 protein positive cells were(10. 22 ± 2. 16) %,(51. 86 ± 4. 05) %,(26. 31 ± 3. 39) %.
