摘  要:目的探讨黄芪多糖对慢性萎缩性胃炎大鼠蛋白酪氨酸激酶1/信号转导子与激活子3(JAK1/STAT3)信号传导通路的影响。方法将85只SPF级健康雄性SD大鼠随机分为正常组13只和造模组72只。造模组用N-甲基-N-硝基-N-亚硝基胍、水杨酸钠、雷尼替丁和饥饱失常四因素综合造模法建立慢性萎缩性胃炎模型;正常组给予普通饲料及无菌过滤水饲养。将模型大鼠随机分为模型组、对照组和低、中、高剂量实验组,每组13只。模型组和正常组均按5 mL·kg-1的剂量灌胃给予0.9%NaCl,qd;低、中、高剂量实验组均按5 mL·kg-1的剂量分别灌胃给予0.1,0.2和0.4 g·mL^-1黄芪多糖溶液,qd;对照组灌胃给予0.2 g·mL^-1维酶素溶液5 mL·kg-1,qd。各组大鼠均给药8周。比较各组的血清胃泌素(GAS)及白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,以及胃黏膜JAK1和STAT3蛋白的表达水平。结果低、高剂量实验组和对照组、模型组、正常组的血清GAS分别为(51.37±4.28),(60.07±2.48),(60.05±2.41),(46.73±5.15)和(63.26±4.63)pg·mL^-1,IL-6分别为(10.49±1.16),(3.28±0.32),(3.25±0.36),(14.64±1.37)和(1.47±0.18)pg·mL^-1,TNF-α分别为(9.47±1.04),(2.49±0.28),(2.44±0.26),(13.58±1.52)和(0.51±0.06)pg·mL^-1,胃黏膜JAK1蛋白相对表达量分别为0.65±0.08,0.33±0.04,0.32±0.03,0.83±0.11和0.14±0.02,STAT3蛋白相对表达量分别为0.61±0.06,0.25±0.03,0.23±0.02,0.94±0.13和0.19±0.02,低、高剂量实验组和对照组、正常组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论黄芪多糖能剂量依赖性地改善慢性萎缩性胃炎大鼠萎缩病变,调节胃肠激素释放,抑制炎症因子释放,其作用可能与抑制JAK1/STAT3信号传导通路活性有关。Objective To analyze the effect of astragalus polysaccharides on protein tyrosine kinase 1/signal transducer and activator 3(JAK1/STAT3) signal transduction pathway in rats with chronic atrophic gastritis. Methods A total of 85 SPF male SD rats were randomly divided into normal group(n=13 rats) and model building group(n=72 rats). The model building group was established chronic atrophic gastritis model by the method of N-methyl-N-nitro-N-nitrosoguanidine, sodium salicylate, ranitidine and anorexia. The normal group received common feed and sterile filtered water. The model rats were randomly divided into model group,control group and experimental-L,-M,-H groups with 13 rats per group. The model and normal groups were given 0. 9% NaCl at the dose of 5 mL·kg^-1 by intragastric administration,qd. The experimental-L,-M,-H groups received 0. 1,0. 2,0. 4 g·mL^-1 astragalus polysaccharides solution at the dose of 5 mL·kg^-1 by intragastric administration,qd. The control group was treated with 0. 2 g·mL^-1 vitacoenzyme solution at the dose of 5 mL ·kg^-1 by intragastric administration,qd. All rats were treated for 8 weeks.The serum levels of gastrin(GAS),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α),and the expression levels of JAK1 and STAT3 protein in gastric mucosa were compared among each group. Results The serum levels of GAS in experimental-L,-H groups and control group,model group,normal group were(51. 37 ± 4. 28),(60. 07 ± 2. 48),(60. 05 ± 2. 41),(46. 73 ± 5. 15) and(63. 26 ± 4. 63) pg·mL^-1,IL-6 were(10. 49 ± 1. 16),(3. 28 ± 0. 32),(3. 25 ± 0. 36),(14. 64 ± 1. 37) and(1. 47 ± 0. 18) pg·mL^-1,TNF-α were(9. 47 ± 1. 04),(2. 49 ± 0. 28),(2. 44 ± 0. 26),(13. 58 ± 1. 52) and(0. 51 ± 0. 06) pg·mL^-1,relative expression levels of JAK1 protein in gastric mucosa were 0. 65 ± 0. 08,0. 33 ± 0. 04,0. 32 ± 0. 03,0. 83 ± 0. 11 and 0. 14 ± 0. 02,relative expression levels of STAT3 protein were 0. 61 ± 0. 06,0. 25 ± 0. 03,0. 23 ± 0. 02,0. 94 ± 0. 13 and 0. 19 ± 0. 02.Compared with mo
