摘  要:目的:利用蝉拟青霉对黄芪进行发酵处理,探究发酵后所得菌质有效成分含量变化及其抗高尿酸血症活性。方法:采用硫酸-苯酚比色法、NaNO 2-Al(NO 3)3比色法、酸碱滴定法对黄芪及黄芪发酵菌质中多糖、总黄酮、总酸的含量进行测定;采用氧嗪酸钾盐诱导高尿酸血症(HUA)大鼠模型,将雄性SD大鼠随机分为对照组、模型组、菌质(150、300 mg·kg^-1)组、黄芪(150、300 mg·kg^-1)组及苯溴马隆(20 mg·kg^-1)组,连续灌胃给药14 d,分别对大鼠血清中的尿酸(UA)、肌酐(CRE)、尿素氮(BUN)、总胆固醇(TC)、三酰甘油(TG)、丙氨酸转氨酶(ALT)及天冬氨酸转氨酶(AST)水平进行测定。结果:黄芪药材经蝉拟青霉双向发酵后,总多糖的含量显著升高,总黄酮的含量显著降低,总酸的含量变化不明显。与对照组比较,模型组大鼠血清中UA、BUN、TC、TG、ALT和AST水平显著升高(P<0.05,P<0.001);与模型组比较,黄芪组及黄芪发酵菌质组大鼠血清中UA、BUN、TC、TG、ALT和AST水平显著降低(P<0.05,P<0.01,P<0.001),且菌质组效果明显优于黄芪组。结论:黄芪经蝉拟青霉发酵后有效成分含量发生较大变化,所得蝉拟青霉/黄芪双向发酵菌质具有良好的抗高尿酸血症作用。Objective:To establish a bidirectional fermentation system of Paecilomyces cicadae/Astragalus membranaceus,and to explore the content change of active ingredients the effect of lowing uric acid in fermentation process.Methods:The contents of polysaccharide,total flavonoids and total acid were determined by sulfuric acid-phenol colorimetry,NaNO 2-Al(NO3)3 colorimetry and acid-base titration method.Male SD rats were randomly divided into 7 groups,control group,model group,fermentation groups with high and low doses,Astragalus membranaceus groups with high and low doses and benzbromarone group.Biochemical technique was used to detect serum UA,BUN,CRE,TC,TG,ALT and AST level.Results:The contents of polysaccharides in Astragalus membranaceus were increased,while flavonoids content reduced.And the content of total acid remain roughly the same.At the same time,UA,BUN,CRE,TC,TG,ALT and AST level in the fermentation groups significantly decreased compared with the model group.That means the bidirectional fermentation system of Paecilomyces cicadae/Astragalus membranaceus possess certain effect of hyperuricemia.Conclusion:Paecilomyces cicadae can ferment Astragalus membranaceus medicinal materials and the yield of effective components changed significantly after fermentation.In addition,it can significantly work on hyperuricemia treatment.
