摘 要:目的建立黄瑞香叶提取物中总黄酮、祖师麻甲素和7-羟基香豆素含量的测定方法。方法采用比色法,以芦丁为对照品,采用亚硝酸钠-硝酸铝-氢氧化钠显色,在500 nm波长处测定吸光度值A并计算总黄酮含量;采用UPLC法,测定祖师麻甲素和7-羟基香豆素的含量。采用BEH C_(18)(50 mm×2.1 mm,1.7μm)色谱柱;流动相:乙腈-甲酸(15∶85);检测波长:327 nm;流速:0.3 mL·min^(-1)。结果芦丁质量浓度在0.06~0.36 mg·mL^(-1)范围内与吸光度值A呈良好的线性关系(r_1=0.999 6),平均回收率为97.56%(RSD=1.19%)(n=6);总黄酮含量均值为44.47%。祖师麻甲素和7-羟基香豆素质量浓度分别在0.08~0.48 (r_2=0.999 9)和0.012 4~0.074 4(r_3=0.999 9)μg·mL^(-1)范围内与吸光度值A呈良好的线性关系;平均回收率分别为97.65%(RSD=1.79%)和97.18%(RSD=1.52%)(n=9);平均含量分别为11.28%和0.54%。结论该法操作简便,结果准确,重复性与精密度良好,方法可靠,可作为黄瑞香叶提取物的含量测定方法。Objective To establish a method for the determination of total flavones,daphnetin and umbelliferone in Daphnes folium.Methods The content of total flavonoids was determined by colorimetry with NaNO 2-Al(NO 3)3-NaOH color-test at the wavelength of 500 nm with the reference of rutin.A UPLC method was used to determine the contents of daphnetin and umbelliferone.The chromatographic column was BEH C 18(50 mm×2.1 mm,1.7μm);the mobile phase was acetonitrile-formic acid(15∶85).The detection wavelength was 327 nm,and the flow rate was 0.3 mL·min-1.Results There was a good linear relationship between the quality concentration of rutin and the absorbance in the range of 0.06-0.36 mg·mL-1(r 1=0.9996).The average recovery was 97.56%(RSD=1.19%)(n=6).The average content of total flavonoids was 44.47%.There was a good linear relationship between the absorbance values of daphnetin and umbelliferone in the ranges of 0.08-0.48(r 2=0.9999)and 0.0124-0.0744μg·mL-1(r 3=0.9999);the average recovery were 97.65%(RSD=1.79%)and 97.18%(RSD=1.52%)(n=9),respectively;the average contents were 11.28%and 0.54%,respectively.Conclusion The method is simple,accurate,reproducible and stable,and can be used for the content determination of the effective extract of Daphnes folium.
