摘　　要：目的建立能同时测定五味健脑合剂中2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、朝藿定A、朝藿定B、朝藿定C、淫羊藿苷、6-姜辣素和8-姜酚含量的一测多评法(QAMS)。方法采用Agilent TC-C_(18)色谱柱(250 mm×4.6 mm,5μm);柱温:30℃;以乙腈-1 mL·L^(-1)磷酸为流动相,梯度洗脱;流速:1.0 mL·min^(-1);检测波长分别为320(检测2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷)和270 nm(检测朝藿定A、朝藿定B、朝藿定C、淫羊藿苷、6-姜辣素和8-姜酚);以淫羊藿苷为内参物,测定2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、朝藿定A、朝藿定B、朝藿定C、6-姜辣素和8-姜酚的相对校正因子(RCF),利用RCF计算指标性成分的含量,并与外标法(ESM)测定结果进行对比,验证QAMS的可行性。结果 7种成分分别在7.68~192.00,2.86~71.50,4.69~117.25,7.87~196.75,5.46~136.50,1.83~45.75和0.67~16.75μg·mL^(-1)范围内线性关系良好(r≥0.999 1),平均回收率(RSD值)分别为99.31%(0.97%),98.50%(1.25%),98.19%(1.35%),100.04%(0.67%),97.99%(1.40%),96.94%(1.29%)和97.26%(0.91%),ESM和QAMS法测定结果无明显差异。结论 QAMS评法操作便捷、结果准确、重复性好,可用于五味健脑合剂中7种成分的同时测定。Objective To establish a quantitative analysis method for the simultaneous determination of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,epimedin A,epimedin B,epimedin C,icariin,6-gingerol and 8-gingerol in Wuwei Jiannao Mixture by multi-components with a single-marker(QAMS).Methods The chromatographic separation was carried out on an Agilent TC-C 18 column(250 mm×4.6 mm,5μm)with the mobile phase consisting of acetonitrile-1 mL·L-1 phosphoric acid aqueous solution with gradient elution at a flow rate of 1.0 mL·min-1.The column temperature was set at 30℃.The detection wavelengthes were set at 320 nm for 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,and 270 nm for epimedin A,epimedin B,epimedin C,icariin,6-gingerol and 8-gingerol.The relative correction factors(RCF)of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside,epimedin A,epimedin B,epimedin C,6-gingerol and 8-gingerol were determined by using icariin as a reference substance.The quantitative results were compared between the external standard method(ESM)and QAMS.Results The method was developed with good linearity over the range of 7.68-192.00,2.86-71.50,4.69-117.25,7.87-196.75,5.46-136.50,1.83-45.75 and 0.67-16.75μg·mL-1(r≥0.9991)of 7 components.The average recovery rates and RSDs were 99.31%(0.97%),98.50%(1.25%),98.19%(1.35%),100.04%(0.67%),97.99%(1.40%),96.94%(1.29%)and 97.26%(0.91%),respectively.There was no significant difference between ESM and QAMS.Conclusion The QAMS method is simple,accurate and reproducible.It can be used for the simultaneous determination of the 7 components in Wuwei Jiannao Mixture.
