止血镇痛胶囊对功能失调性子宫出血大鼠的治疗作用

摘　　要：目的研究止血镇痛胶囊对大鼠功能性子宫出血的治疗作用。方法将受精雌鼠分为空白对照组、模型对照组、卡巴克络对照组、独一味软胶囊组及止血镇痛胶囊低、中、高剂量组。妊娠第7天时除空白对照组外,其余各组给予米非司酮和米索前列醇造成不完全流产。第8~14 d各给药组灌胃给予相应药物,空白对照组和模型对照组灌胃生理盐水。7~14 d期间,每日分别于8∶00和20∶00采集阴道出血量。第15天时取血,Elisa测试血清中雌二醇(E2)和孕酮(P)的含量,并在光镜下观察大鼠子宫组织病理变化。结果与模型对照组比较,止血镇痛胶囊低剂量组子宫出血时间、子宫出血总量、给药第7天的子宫出血量显著减少(P<0.05);止血镇痛胶囊中剂量组大鼠血清中E2含量显著升高(P<0.01),止血镇痛胶囊高剂量组大鼠血清中E2和P含量均显著升高(P<0.01),止血镇痛胶囊3个剂量组大鼠子宫中绒毛及蜕膜细胞残留均显著减少(P<0.05)。结论止血镇痛胶囊能减少子宫出血时间,提高激素水平,促进子宫绒毛及蜕膜细胞残留排出,对大鼠功能性子宫出血具有明显的改善作用。Objective To study the therapeutic effect of Zhixue Zhentong Capsules on functional uterine bleeding in rats.Methods The fertilized female rats were divided into blank control group,model control group,carbazochrome control group,Duyiwei Soft Capsules group,as well as Zhixue Zhentong Capsules groups with low,medium and high doses.On the 7th day of pregnancy,except for the blank group,all the other groups were given mifepristone and misoprostol to induce incomplete abortion.During the 8th-14th days,the blank control group and the model control group were orally administered with normal saline while other groups were given corresponding drugs by gavage administration.During the 7th-14th day,vaginal blood was collected respectively at 8∶00 and 20∶00.On the 15th day,the blood was taken,in which the serum levels of estradiol(E2)and progesterone(P)were measured by Elisa.Meanwhile,the pathological changes of rat uterus were observed under light microscope.Results Compared with the control group and the model group,uterine bleeding days,the total amount of uterine bleeding,and the amount of uterine bleeding on the 7th day of administration were significantly decreased in the low-dose group of Zhixue Zhengtong Capsules(P<0.05).The level of serum E2 in the medium dose group was significantly increased(P<0.01),and level of the serum E2 and progesterone in the high-dose group of Zhixue Zhentong Capsules were significantly increased(P<0.01).Residues of villus and decidual cells in the uterus of the 3 dose groups were significantly decreased(P<0.05).Conclusion Zhixue Zhentong Capsule can reduce the time of uterine bleeding,increase hormone levels,promote the release of uterine villi and decidual cells,thus significantly improve the functional uterine bleeding in rats,which provides references for clinical application.
