摘 要:目的:研究藏族精子蛋白表达水平与汉族的差异及其对高原缺氧环境适应的意义。方法:收集移居汉族、世居藏族和平原汉族可育男性精液标本。纯化精子进行差异蛋白质组学分析,揭示不同组别蛋白表达水平差异,筛选高原环境中藏、汉精子差异表达蛋白,分析其参与的通路与生物学过程,揭示其对高原环境适应的意义。结果:移居汉族和世居藏族精子差异表达蛋白富集于凋亡相关的分子通路,而移居汉族与平原汉族的精子差异表达蛋白在DNA损伤修复相关通路中富集。蛋白质组分析显示移居汉族精子中膜联蛋白表达上调,但免疫印迹未能验证其在移居汉族与世居藏族精子中的差异。结论:高原低氧环境导致精子DNA损伤。移居汉族与世居藏族精子膜联蛋白差异表达提示汉族和藏族精子通过不同机制适应高原低氧环境,维持基因组完整性。Objective To study the difference of sperm protein expression between Tibetan and Han Chinese and its significance in adaptation to high altitude hypoxia environment.Methods Semen specimens were collected from immigrated Han Chinese,native Tibetans,and plain Han Chinese.The purified sperm specimens were analyzed by differential proteomics to reveal the differences of protein expression levels in different groups.The differential expression proteins of Tibetans and Han Chinese sperm in high altitude environment were screened,and the pathways and biological processes involved in them were analyzed to reveal the significance of their adaptation to high altitude environment.Results The differentially expressed proteins in the spermatozoa of the Han Chinese and native Tibetans were enriched in the apoptotic related molecular pathways.However,the differentially expressed proteins in the spermatozoa of the immigrated Han Chinese and the plain Han Chinese are enriched in the DNA damage repair related pathways.Proteomic analysis showed that the expression of Annexin was up-regulated in the sperm of Han Chinese and native Tibetans,but Western blot could not verify the difference between the two groups.Conclusions High altitude hypoxic environment causes DNA damage in sperm.The difference of Annexin expression between Han Chinese and Tibetans spermatozoa indicate that Han Chinese and Tibetans sperm adapt to high altitude hypoxia environment and maintain genomic integrity through different mechanisms.
