摘  要:目的:探讨大鼠脊髓损伤(spinal cord injury,SCI)后神经干细胞(neural stem cells,MSCs)移植与补阳还五汤(Buyang Huanwu decoction,BYHWD)对大鼠脊髓全横断损伤的修复。方法:对脊髓损伤的大鼠分别进行鼠源MSCs移植或补阳还五汤治疗及两者联合治疗,观察治疗后大鼠行为学变化;检测各组细胞wnt3a与β-actin比值。结果:t检验分析表明,MSCs+BYHWD组大鼠的BBB评分值高于脊髓损伤组[(17.77±0.97)vs(15.33±1.02),P<0.05];Western-Blot结果表明,与对照组相比,MSCs组、BYHWD组及MSCs+BYHWD组的wnt3a表达上调,且MSCs+BYHWD组远高于MSCs组和BYHWD组。结论:补阳还五汤辅助MSCs移植治疗SD大鼠脊髓损伤有一定改善效果,且改善效果远高于MSCs或BYHWD单药治疗。Objective:To investigate the effect of neural stem cell(MSCs)transplantation and Buyang Huanwu decoction(BYHWD)on spinal cord injury(SCI)in rats.Methods:The rats with spinal cord injury were treated with rat-derived MSCs transplantation or Buyang Huanwu decoction and the combination of the two therapies.The behavioral changes of the rats were observed and the ratios of wnt3a toβ-actin in each group of cells were detected.Results:The student’s t tests showed that the BBB score of the rats was higher in the MSCs+BYHWD group than the SCI group[(17.77±0.97)vs(15.33±1.02),P<0.05].Western-Blotting results showed that the expression of wnt3a in the MSCs group,BYHWD group and MSCs+BYHWD group was up-regulated as compared with the control group and furthermore its expression was much higher in the MSCs+BYHWD group than the MSCs group and BYHWD group.Conclusion:Buyang Huanwu decoction-assisted MSCs transplantation for the treatment of SCI in rats has certain improvement effect and its effect is much higher compared with MSCs and BYHWD monotherapy.
