摘  要:在海南省临高县发现一种淮山叶部炭疽病病害,明确其病害种类及开展生物学特性研究。通过病原分离、纯化、显微镜镜检、致病性测定,对代表性病原菌株LGDY03结合形态学用MEGA6.0软件ML方法构建基于ITS、ACT、CHS-1、BT2和GAPDH多基因序列构建的同源性系统发育树。明确该菌株为炭疽菌Colletotrichum alatae,气生菌丝绒毛状,菌落初为白色,后变为灰色再到深灰色,有橘红色孢子团的产生。分生孢子无色,整体长椭圆形,单胞,大小为(12.5~22.5)μm×(3.5~6)μm。其生物学特性研究表明,最适培养基为PDA,最适温度为28℃,最适pH值为7,最适碳源为麦芽糖,最适氮源为甘氨酸,光照对菌丝生长无显著性影响。A leaf anthrax of yam disease was found in Lingao County,Hainan Province,and its pathogen and biological characteristics were identified.Through pathogen isolation,purification,microscopy,pathogenicity determination,the representative pathogenic bacteria strain LGDY03 was identified by morphologic characteristics and multi-locus phylogeny analysis.The homology phylogenetic tree based on ITS,ACT,CHS-1,BT2,GAPDH multigene sequences constructed by MEGA6.0 software ML.The pathogen was identified as Colletotrichum alatae,and aerial mycelium was fluffy.It was white at the initial stage,and then changed to dark gray with orange spore piles.The conidia were colorless,long elliptic and unicellular,with the size of(12.5~22.5)μm×(3.5~6)μm.The research on its biological characteristics showed that:the optimum nutrient medium was PDA;the optimum temperature was 28℃;the optimum pH value was 7;the optimum carbon source was maltose;and the optimum nitrogen source was glycine.Light had no significant effect on the mycelial growth.
