摘 要:目的探讨单侧前庭功能丧失大鼠中枢代偿的机制。方法将45只成年健康SD大鼠分为空白对照组(9只)、假手术对照组(18只)、模型组(18只)。空白对照组正常饲养,假手术对照组鼓室内一次性注射生理盐水,模型组鼓室内一次性注射氯仿,注射后1 h进行模型评价。造模成功3 d后,选择免疫组化法、反转录聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)及Western-blot法观察实验耳侧前庭内侧核、小脑绒球中组胺受体H1、H2、H3亚型的表达。结果假手术对照组与空白对照组前庭内侧核、小脑绒球中组胺受体H1、H2、H3亚型含量比较,差异无统计学意义(P>0.05),模型组与假手术对照组、空白对照组比较表达明显上调(P<0.05)。模型组前庭内侧核与小脑绒球组胺受体H1、H2、H3亚型含量比较,差异无统计学意义(P>0.05)。结论单侧前庭功能丧失可导致患侧前庭内侧核、小脑绒球中组胺受体H1、H2、H3亚型表达上调,是前庭代偿的可能机制,将为今后防治前庭神经损伤及促进前庭功能代偿提供新思路。Objective To investigate the mechanism of central compensation in rats with unilateral vestibular dysfunction.Methods Forty-five adult healthy SD rats were randomly divided into blank control group(n=9),sham operation control group(n=18)and model group(n=18).The blank control group was fed normally,the sham operation control group was injected with normal saline once,and the model group was injected with chloroform once in tympanic cavity.The model was evaluated 1 h after injection.After three days,the model group was established,immunohistochemistry,PCR and Westernblot were used to observe the expression of histamine receptor H1,H2 and H3 in the medial vestibular nucleus and cerebellar choriocyte.Results The expression of histamine receptor H1,H2,H3 subtypes in medial vestibular nucleus and cerebellar choriocyte in the sham operated control group was not significantly different from that in the blank control group(P>0.05).The expression of histamine receptor H1,H2 and H3 subtypes in the model group was significantly up-regulated compared with the sham operation control group and the blank control group(P<0.05).There was no significant difference between the medial vestibular nucleus and cerebellar choriocyte(P>0.05).Conclusions Unilateral vestibular dysfunction can lead to upregulation of histamine receptor H1,H2,H3 subtypes in the medial vestibular nucleus and cerebellar choriocyte,which may be a possible mechanism of vestibular compensation,and provide a new idea for prevention and treatment of vestibular nerve injury and promotion of vestibular function compensation in the future.
