摘 要:首次建立了超高效合相色谱法(Ultra Performance Convergence Chromatography,UPC2)快速检测大黄中5种蒽醌类化合物的方法。大黄样品用甲醇提取后,以CO2和甲醇为流动相和改性剂,梯度洗脱,色谱柱为HSS SB C18,柱温40℃,流速2.0 mL/min,进样量1.0μL,检测波长260 nm;整个分析过程仅需1.6 min。该方法的线性范围除大黄酸为1.0~100.0 mg/kg外,其余均为0.5~100.0 mg/kg,日内及日间精密度的RSD范围分别为1.8%~3.5%和0.9%~1.2%。在最优条件下,对10批不同产地的大黄进行检测。结果表明,UPC2方法具高效快速、操作简单、试验成本低等优点,可以满足大黄中5种蒽醌类化合物的快速检测。Based on Ultra performance convergence chromatography,sub-2μm particles were used as column.The retention behavior of anthraquinones represented by emodin and its related substances under different separation conditions was studied for the first time,and the rapid separation of five anthraquinones in rhubarb samples was realized.In the tested stationary phase,using carbon dioxide as the mobile phase,methanol as the cosolvent completed the rapid separation of the target in the rhubarb sample within 1.6 min,and the method was verified.And the method verification was carried out.The linear range of the method was in the range of 1.0~100.0 mg/kg,the others was 0.5~100.0 mg/kg,and the RSD ranges of the intra-day and the inter-day precision were between 1.8%~3.5%and 0.9%~1.2%,respectively.The results showed that the method of UPC^2 had the advantages of high efficiency,simple operation and low experimental cost,and can be used for the rapid detection of five anthraquinones in rhubarb.
