摘 要:目的建立多波长HPLC结合化学模式识别技术,比较漳州陈皮不同炮制品的质量。方法采用多波长高效液相法(MW-HPLC),按时间波长程序283 nm(0~6 min)、283 nm(6~19 min)和334 nm(19~30 min)同时测定不同漳州陈皮炮制品中橙皮苷、橙皮素和川陈皮素含量。应用化学模式识别技术中聚类分析(HCA)、主成分分析(PCA)和偏最小二乘法-判别分析(PLS-DA),对不同陈皮炮制品质量进行分析评价。结果所测漳州陈皮样本中黄酮类成分的含量从高到低依次为:四制陈皮>蒸陈皮>麸炒陈皮>炒陈皮>醋陈皮>陈皮生品>蜜陈皮>土炒陈皮>盐陈皮;HCA和PCA分析结果一致,9种炮制品可分为3大类,其中生品1类,四制陈皮1类,其余炮制品1类;PLS-DA分析结果表明,橙皮苷、川陈皮素和橙皮素等6个成分是主要差异性标志物。结论不同炮制方法陈皮质量有差异,漳州四制陈皮的黄酮类活性成分含量高,且检测出橙皮素成分,具有很好的品质特色。建立的含量测定结合化学模式识别方法可用于漳州陈皮炮制品的综合质量评价。Objective To comparative analyze the quality of different processed Citri Reticulatae pericarpium(CRP)from Zhangzhou by multi-wavelength HPLC(MW-HPLC)combined with chemical pattern recognition technology.Method The contents of hesperidin,hesperetin and nobiletin in different processed CRP were determined by MW-HPLC,which was programmed at 283 nm(0-6 min),283 nm(6-19 min)and 334 nm(19-30 min)according to the time wavelength.The quality of different processed CRP was evaluated by chemical pattern recognition including hierarchical cluster analysis(HCA),principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA).Results The contents of flavonoids in all of the tested CRP samples from high to low was four made CRP,steamed CRP,bran fried CRP,fried CRP,vinegar made CRP,raw CRP,honey made CRP,soil fried CRP and salt made CRP.Nine kinds of processed CRP were divided into 3 categories,including raw CRP,four made CRP and other processed CRP through HCA and PCA analysis.The results of PLS-DA analysis showed that six components including hesperidin,hesperetin and nobiletin were the main difference markers.Conclusion The quality of CRP was different in different processing methods.Among of them,the content of flavonoids was high and the component of hesperitin was detected in Zhangzhou four made CRP with good quality characteristics.The method of content determination based on MW-HLPC combined with chemical pattern recognition could be used for the comprehensive quality evaluation of processed CRP products.
